摘 要:目的:利用代谢组学技术,研究苍耳子及其配伍黄芪后,大鼠肝脏组织病理、血浆生化指标、尿液代谢图谱的变化,探讨黄芪配伍苍耳子对肝脏的减毒作用。方法:大鼠30只随机分为正常对照组、苍耳子组(21.0g/kg)、黄芪配伍组(31.5g/kg),灌胃给药,连续给药28d,收集大鼠24h尿液,测定尿液UPLC-TOF-MS图谱;肝门取血做生化检测;取肝脏做组织病理学检查。结果:给药28d,苍耳子组及黄芪配伍组较对照组ALT、AST均升高,肝组织出现病理改变;黄芪配伍组较苍耳子组ALT、AST降低,肝组织病变明显减轻。苍耳子组、黄芪配伍组尿液代谢物表型的聚类分布均偏离对照组,但黄芪配伍组尿液代谢物表型的聚类分布位于对照组与苍耳子组之间。结论:苍耳子(21.0g/kg)长期给药可造成肝功能的明显损害,配伍黄芪后(31.5g/kg)具有一定的减毒作用。To study the changes of liver tissue pathology, serum biochemical indicators and urine metabonomic profiling of rats in Fruc- tus Xanthii group and compatibility group of Radix Astragali by metabonomic technology,in order to explore whether Radix Astragali could de- crease the hepatoxicity of Fructus Xanthii. Methods : Thirty male Wistar rats were divided into the formula with Radix Astragali group, the Fructus Xanthii group and the control group by random-digit table method, each group comprised 10 rats. The rats in the Fructus Xanthii group were gavaged with the suspension of cocklebur fruit which contained Fructus Xanthii crude drug 21.0g/kg, rats in the formula group ga- vaged with the suspension of cocklebur fruit and milkvetch root having Fructus Xanthii crude drug 21.0g/kg and Radix Astragali crude 10. 5g/kg, and once daily for 28 days. The rats in the control group were gavaged with same volume of 0.9% sodium chloride injection once dai- ly for 28 days. The urine samples over 24 hours of each rat were collected and the metabolic profiles were determined by ultra-high perform- ance liquid chromatography/time of flight mass spectrometry, blood taken from hepatic portal vein, the serum biochemical indicators were measured, Subsequently,the rats were sacrificed, the liver histopathology was graded. Results: After administrated for 28 d, the levels of ALT, AST of rats in the Fructus Xanthii group and compatibility group of Radix Astragali were markedly higher than those in the control group and the liver tissue appeared pathological changes. Nevertheless, compared with the Fructus Xanthii group, those of the compatibility group of Radix Astragali reduced ~md the lesions of liver arised apparent ease. The distributions of rats'urine metabolite phenotypic clustering in drug groups were deviated from the control group, but that of the compatibility group of Radix Astragali lied in between the control group and the Fructus Xanthii group.
