摘 要:目的:观察全蚕粉对2型糖尿病胰岛素受体信号传导通路的影响,探讨全蚕粉逆转2型糖尿病胰岛素抵抗的分子作用机理。方法:以高热量饲料喂养大鼠8周加小剂量链脲佐菌素(STZ,25mg/kg)腹腔注射建立2型糖尿病大鼠模型。应用全蚕粉进行干预,以阿卡波糖片作为阳性对照药,用Real-time PCR和Western blotting技术,观察全蚕粉对Akt,InsR,PI3K基因及其蛋白表达的影响。结果:与模型组比较,全蚕粉组0.5、1.0、2.0g/kg肝组织Akt,InsR,PI3K mRNA的相对表达量无显著性差异,肝组织Akt,InsR,PI3K蛋白表达量显著上调(P﹤0.05)。结论:全蚕粉对2型糖尿病大鼠的影响可能在翻译水平上通过PI3K信号传导途径增强胰岛素生物效应,从而逆转2型糖尿病大鼠胰岛素抵抗。To study the effects of Silkworm Powder(SP) on glucose and lipid metabolism in Type 2 Diabetic rats and exam the effects of Silkworm powder on PI3K/AKT signaling pathway in Type 2 Diabetic rats. In order to understand the mechanisms of SP'protective effects on Type 2 Diabetesmellious. Methods: Type 2 Diabetic model was induced. Rats were randomly divided into five groups. To investigate the effects of SP on PI3K/AKT signaling pathway in hepatic tissue by real time-PCR and Western blotting after 6 weeks of intragastric administra- tion with vehicle. Results: Compared with type 2 DM groups, no significant changs in the mRNA level of Akt, InsR, PI3K in all treatment groups . Tile protein expl ession of Akt in heptic tissue significantly increased in middle and high dose of groups ;The protein expression of In- sR and PI3K in hepatic tissue also significantly increased( P 〈 0.05). Conclusion: The results of present study demonstrated that SP can ef- fectively improve the gly-lipid metabolim and have an improvement on IR. Silkworm powder can improve PI3K/AKT signaling pathway in Type 2 Diabetic rats .
