糖心安对糖尿病动物感觉神经的影响

摘　　要：目的:评价糖心安胶囊对糖尿病神经病变的防治作用。方法:雌性小鼠一次性腹腔注射链脲佐菌素150mg/kg以复制糖尿病模型,将糖尿病小鼠随机分为模型组、卡马西平0.1 g/kg、糖心安0.45 g/kg、糖心安0.15 g/kg、糖心安0.05 g/kg。灌胃给药,每天一次,连续14天,模型组和正常对照组分别给于等体积0.5%羧甲基纤维素钠,于末次给药后1小时测定其热痛阈值、压痛阈值、血糖水平、Aδ纤维以及C纤维敏感性等指标。结果:Ⅰ型糖尿病小鼠热痛阈值、机械刺激痛阈值降低,在CS条件下(30V),C纤维对电刺激产生明显高反应性;在post-CS条件下(15V),Aδ纤维在强电刺激后对低电刺激的易化(facilitation)程度明显降低;糖心安0.15 g/kg、0.45 g/kg可显著增加糖尿病小鼠热刺激以及机械刺激疼痛的阈值,糖心安各剂量组降低糖尿病小鼠C纤维在强电压刺激下的高敏感性,糖心安0.15 g/kg、0.45 g/kg可改善Aδ纤维在强电刺激后易化程度。结论:糖心安改善糖尿病神经病变所产生的疼痛症状可能与其降低糖尿病小鼠C纤维在强电压刺激下的高敏感性,以及改善Aδ纤维在强电刺激后易化程度有关,但与血糖水平无明显关系,其详细作用机理还有待能进一步研究。Evaluation of the effect of Tangxinan on diabetic neuropathy（DN）. Methods： Mice were induced DN by ip STZ 150 mg/kg. 3 days later, DN mice were divided into 5 groups. These mice were ig Tangxinan 450mg/kg, 150mg/kg, 50mg/kg, carbamazepine 100mg/kg or 0.5% same volume of CMC respectively for 14 days. The hot plate test and toe pinch test were used to measure the threshold of pain in DN mice, and the sensibility of A~ and C nerve fibers. Results ： The threshold of pain were significantly decreased, wind-up of C-fiber-mediated withdrawal reflex to electricity- stimulation produce significantly higher reactivity under the CS condition （30V）and the facilitation of A^-fiber- mediated withdrawal reflex was smaller under post-CS condition（15V） in DN mice. Tangxinan can significantly increase the threshold of pain, ameliorate the higher reactivity of C-fiber-mediated withdrawal reflex and the facilitation of A-fibe-mediated withdrawal reflex. Con- clusions： Tangxinan can reduce the sensitivity of pain in diabetic animals, suggesting that angxinan has a protective effect on the diabetic neuropathy. These actions were not related to decrease the level of blood glucose.
