摘  要:目的:研究槐定碱对内毒素(LPS)致急性肺损伤(ALI)小鼠血清中白介素6(IL-6)、白介素10(IL-10)、一氧化氮(NO)、超氧化物歧化酶(SOD)及丙二醛(MDA)含量的影响。方法:随机将60只KM小鼠分成6组:①正常组、②模型组、③槐定碱5mg/kg预防给药组、④槐定碱2.5mg/kg组、⑤槐定碱5mg/kg组、⑥槐定碱9 mg/kg组,每组10只。①、②、④、⑤、⑥组腹腔注射生理盐水0.5ml,连续6d。③组腹腔注射5mg/kg槐定碱,连续6d。上述②~⑥组均在末次注射生理盐水或药物后1h腹腔注射LPS 9mg/kg造模,造模2h后④~⑥组分别给槐定碱2.5mg/kg、5mg/kg、9mg/kg,4h后取材。结果:槐定碱组IL-6较模型组降低(P<0.05),槐定碱组IL-10含量介于正常组与模型组之间(P<0.01),NO、SOD、MDA结果显示:与模型组比较,给药四组氧化损伤均有明显改善,其中5mg/kg的槐定碱组效果较好。病理切片显示,模型组小鼠肺组织结构明显遭到破坏,槐定碱组较模型组有不同程度的改善。结论:槐定碱能降低ALI小鼠血清中IL-6、IL-10、NO、MDA的含量,提高SOD含量,改善肺脏病理损伤,具有抗ALI的作用。To study the effect of serum IL-6, IL-10, NO, SOD, MDA of sophoridine in acute lung injury (ALl) mice induced by li- popolysaceharide. Methods: 60 KM mice were randomly divided into 6 groups: normal group, model group, prevention treatment group, so- phoridine high dose group, sophoridine middle dose group, sophorldine low dose group. Each group has 10 mice. The normal gruop, model group,sophoridine high,middle and low groups were lavaged 0.5 ml sodium chloride and the prevention treatment group was lavaged 0.5ml sophoridine every day. It was lasting 6 days. Using disposable intraperitoneal injection of endotoxin method to establish intends to acute lung injury model in mice. The model was bulit in the sixth day by injecting LPS 9 mg/kg. 2 hours later, sophoridine high, middle and low groups were given 9 mg,/kg, 5 mg/kg, 2.5 mg/kg respectively. Using enzyme-linked immunosorbent assay(ELISA) to study the serum IL-6 and IL-10 levels. According to the manual of kits, NO, SOD, MDA were detected and then pethological slides were made. Results: Model group IL-6 content (480.06±73.66) compared to the normal group (225.61±32.89) increased significantly ( P 〈 0.01 ). The value of 3 so- phoridine groups compared to that of model group reduced. (P 〈 0.05 ). Normal group IL-I 0 (10.52±3.43 ) content was low, ILl0 content in others administered drugs groups began to rise after the treatment for 4h and they were higher than normal value but lower than that of mod- el group ( 148.78±24.11 ) . To the value of NO, MDA, the model group was significantly higher than others groups and the SOD content of model group was obviously lower than those of others groups. As to the onteome of pathological slides, the administered drugs groups were improved compared to the model group. Conclusions: Sophoridine can reduce ALl model mice serum IL-6, IL-10, NO, MDA content and in- crease the value of SOD. At the same time, sophoridine can improve pathological lung.
