摘 要:目的:观察大黄分别与甘草、炙甘草、黄连配伍后,以及在复方制剂泻心汤中大黄对大鼠的肝肾毒性,为临床合理应用大黄提供依据。方法 :各配伍组制备总提取物,以大黄生药量计,按18g/kg(大黄生药)灌胃,每天给药1次,连续30天。结果:大鼠在给药大黄总提物18g/kg(大黄生药)两周后就表现出一定肝肾毒性,生化指标ALT、AST、CR呈不同程度地升高。并且给药30天后ALT、AST、CR不同程度地升高,BUN略降低,肝脏有轻度嗜酸性病变及轻度水肿,肾脏出现轻度淤血、水肿和核固缩。除炙甘草外,大黄与甘草、黄连配伍后及在泻心汤中均显示大黄肝肾毒性有所降低。结论:大黄配伍后在一定程度上减轻其肝肾毒性。To investigate the toxicity of rhubarb on kidney and liver of rats among itself and its compatibilities with liquorice root, pre- pared liauorice root, Chinese goldthread rhizome and baikal skullcap root in order to guide rational medication in clinic. Methods: Total ex- tracts of rhubarb and its compatibilities were administered to rats at oral doses of 18g/kg (crude drug) once per day up to 30 days. Results: Toxity of rhubarb on kidney and liver of rats occurred after administration for two weeks. Besides, there were different rises in serum biochem- istry including activities of ALT, AST and CR. After administration for 30 days, activities of ALT, AST and CR rise differently while the con- centration of BUN decreased, and the liver of rats from the rhubarb group showed slightly aeidophilia and edema. Besides, the kidney of rats showed little congestion, edema and karyopyknosis. However, the toxicity of rhubarb on kidney and liver of rats was reduced in its eompati- bilities except for its compatibilities with prepared liauorice root. Conclusion: The toxicity of rhubarb on kidney and liver of rats could be re- duced in its eompatibilities.
