摘　　要：目的:观察虎杖提取物对人外周血单个核细胞炎症因子基因的影响,以期阐明虎杖提取物抗痛风性关节炎的作用机制。方法:分离与培养人外周血单个核细胞,台盼蓝测细胞活力为95%以上,RT-PCR检测人外周血单个核细胞IL-1βmRNA、IL-8mRNA的表达。结果:虎杖提取物1000mg/L,100mg/L组IL-1βmRNA和IL-8 mRNA表达与空白对照组比较有显著性差异(P﹤0.01)。地塞米松磷酸钠组表达与空白对照组比较有显著性差异(P﹤0.01)。结论:虎杖提取物能抑制人外周血单核细胞炎症因子基因的表达,可能是其治疗急性痛风性关节炎的作用机制之一。To observation of polygonum cusPidatnm extract on inflammatory factors expression in human peripheral blood mononuclear cells,in order to clarify the mechanism of Polygonum cuspidatum extract effect on gouty arthritis. Method： solation and cuhure of human pe-ripheral blood mononuclear cells, RT - PCR detection of expression of IL-1βmRNA and I1-8mRNA in peripheral blood mononuelear cell. Re- suits： Polygonum cuspidatum extract 1000rag/L, 100mg/L group inhibited expression of IL-1βmRNA and L-8 mRNA comparison with model group （ p 〈 0.01 ）. Dexamethasone sodium phosphate groups expressed has significant differences compared with the blank control group（ p 〈 0.01 ）. Conclusion： Polygonum cuspidatum extract could inhibit gene expression of inflammatory cytokine in human peripheral blood mononuclear cells may be one mechanisms of treatment its acute gouty arthritis.
