加味圣愈汤调经作用及机制研究

摘　　要：目的:探讨加味圣愈汤的调经作用及其机制。方法:大鼠去卵巢模型、雄激素致无排卵大鼠(ASR)模型灌胃加味圣愈汤1.06、0.53、0.265g/kg分别连续5周和8周,测定子宫、卵巢湿重并行HE染色观察组织形态学变化,放射免疫法测定去卵巢大鼠血清E2、P水平和ASR大鼠血清FSH、LH、E2、P水平;子宫平滑肌试验观察加味圣愈汤含药血清对大鼠离体子宫平滑肌收缩活动和加味圣愈汤十二指肠给药对家兔大体子宫平滑肌活动的影响,BL-420E生物机能系统记录活动曲线。结果:去卵巢大鼠和ASR大鼠子宫、卵巢湿重和系数均下降,两个模型大鼠子宫均有萎缩现象,且ASR大鼠卵巢卵泡发育迟缓,去卵巢大鼠血清E2、P水平及ASR大鼠血清E2、P、FSH水平均明显低于假手术组或溶剂对照组。与模型对照组比较,加味圣愈汤0.53、1.06g/kg能增加两个模型大鼠的子宫、卵巢湿重或脏器系数,1.06g/kg对子宫萎缩和ASR大鼠的卵巢卵泡发育抑制有改善作用;1.06g/kg能升高去卵巢大鼠血清E2水平及ASR大鼠血清LH水平,3个剂量对ASR大鼠血清FSH水平均有明显升高作用;50、100μl含药血清对大鼠离体子宫平滑肌收缩和加味圣愈汤1.06g/kg经十二指肠给药对家兔在体子宫平滑肌收缩均有促进作用,可提高其收缩频率、强度及活动力,与催产素无明显协同作用。结论:加味圣愈汤具有调经作用,灌胃模型大鼠0.265g/kg即起效,在0.265～1.06g/kg内呈较好的量-效正相关;作用机制与调节下丘脑-垂体-卵巢轴内分泌功能和雌激素样作用有关。Objective：To study the effect of regulating menstruation and underlying mechanism for modified Shengyu Decoction.Methods： Animal models of female castrated rats and androgen-induced sterile rats（ASR） were established and then given modified Shengyu Decoction at the doses of 1.06,0.53,0.265g/kg by gavage for 5 weeks and 8 weeks respectively.In vitro uterine muscle strips of rats,and in vivo uterine muslce of rabbits were applied to be given Shengyu Decoction medicated serum of rats or given the drug respectively.Uterine weight and coefficients of the two models were measured,as well as ovarian weight and coefficients of ASR.Then they were fixed by 10% formaldehyde for histopathologic examination by HE staining.Serum levels of estradiol（E2）,progesterone（P） of castrated rats and E2,P,follicle-stimulating hormone（FSH）,luteinizing hormone（LH） of ASR were detected by radio-immunity assay.Curves of uterine muscle contraction activities in vitro and in vivo were recorded with BL-420 biological functional system.Results： Compared with the normal rats,the uterine weight and coefficient of female castrated rats and weight and coefficents of both uterus and ovary in ASR were significantly decreased accompanying uterine atrophy and growth retardation of ovarian follicles.Compared with the model groups,modified Shengyu Decoction significantly increased weight and coefficents of uterus or ovary for the two models and improved histopathologic changes.Also,modified Shengyu Decotion remarkably increased serum level of E2 in female castrated rats and serum levels of E2,P,FSH in ASR.The modified Shengyu Decoction medicated serum of rats significantly increased frequency,magnitude,and motoricity of rat in vitro uterine muscle contraction and the drug itself also showed significantly effects on strengthening magnitude and motoricity for in vivo uterine muscle contraction of rabbits.No obvious synergistic effect between the drug and oxytocin were observed.Conclusion： Modified Shengyu Decoction has effect of
