小檗碱对AGS细胞增殖和IL-8含量的影响

摘　　要：目的:观察小檗碱对胃癌AGS细胞增殖的影响及其对IL-8含量的影响。方法:采用MTT法测定小檗碱及信号通路抑制剂对AGS细胞生长的影响;ELISA法测定小檗碱及抑制剂对AGS细胞IL-8含量的影响。结果:小檗碱对AGS细胞的生长具有良好的抑制作用,该作用呈剂量和时间依赖性,作用AGS细胞48h的IC50值为83μM。进一步研究发现,SB203580、JAK inhibitor和LY2940002能显著抑制AGS细胞的增殖,经SB203580、JAK inhibitor和LY2940002预处理AGS细胞能显著增加小檗碱抑制AGS细胞的增殖作用,而醋竹桃霉素及预处理后加小檗碱干预对AGS细胞的增殖无明显影响。ELISA结果表明,小檗碱(60μM)能够显著降低AGS细胞IL-8含量,而SB203580、JAK inhibitor、姜黄素对AGS细胞IL-8的表达无明显影响;而LY294002能显著降低AGS细胞IL-8含量,经LY294002预孵育0.5 h后小檗碱干预96 h后,AGS细胞中IL-8的含量较两药单用低。结论:小檗碱(60μM)能显著抑制AGS细胞的增殖和IL-8的表达,其作用机制可能与PI3K信号通路的抑制密切相关,而p38 MAPK和JAK信号通路在小檗碱抑制AGS细胞增殖和IL-8表达的作用机制中担当的作用尚待进一步研究和证实。此外,AGS细胞中IL-8的表达与AGS细胞的增殖之间无直接必然的联系。Objective： To investigate the effects of berberine on cell growth and IL-8 level of AGS cells.Methods： The inhibitory effect of berberine on AGS cell growth was measured by MTT method.IL-8 content was measured by ELISA method.Results： Berberine showed significantly inhibitory effect on growth of AGS cells.It inhibited proliferation in a dose-and time-dependent manner,and the IC50 was 83 μM after treatment with berberine for 48 h.To further investigation,we found that LY294002,SB203580 and JAK inhibitor could inhibit the proliferation of AGS cells,and the growth inhibitory effect of berberine on AGS cells was strenghed by LY294002,SB203580 and JAK inhibitor.But was not affected by troleandomycin.Results from ELISA showed that the content of IL-8 was significantly decreased by berberine in AGS cells.But this effect of berberine on IL-8 content could not be affected by SB203580、JAK inhibitor、curcumin.However,LY294002 could also decrease IL-8 level.After pre-incubation with LY294002,the content of IL-8 was lower than those cells treated with LY294002 or berberine alone.Conclusions： Berberine could significantly inhibit the proliferation of AGS cells and IL-8 expression,and its mechanism may be related to inhibition of PI3K signaling pathway.However,the role of p38 MAPK and JAK signal pathways remains to be further studied and confirmed.In addition,there was no necessary relation between cell proliferation and IL-8 expression in AGS cells.
