摘 要:目的:观察雷公藤多苷对结肠炎小鼠外周血CD40/CD40L表达的影响。方法:小鼠随机分成正常组、模型组、雷公藤多苷灌肠、灌胃组及柳氮磺吡啶组,除正常组外,其它动物予TNBS/乙醇法复制溃疡性结肠炎动物模型,雷公藤多苷灌肠、灌胃组均给予20mg/kg,柳氮磺吡啶60mg/kg,正常组及模型对照组均予等容量生理盐水灌肠。通过观察小鼠体重、结肠湿重、结肠重量指数及病理变化和病理损伤评分评价雷公藤多苷治疗小鼠结肠炎的疗效,同时采用流式细胞术检测其外周血CD40/CD40L表达,酶联免疫法检测结肠组织匀浆IL-1β和IL-4水平。结果:与模型组比较,20mg/kg雷公藤多苷灌肠、灌胃组小鼠结肠湿重和结肠重量指数结肠病理损伤评分明显低于模型组,且均有显著性统计学意义(P<0.05或P<0.01)。而且治疗组形态学可见黏膜上皮完整或少量受损,炎细胞散在性分布,基底层可见少量纤维结缔组织增生,溃疡修复。灌肠、灌胃及柳氮磺吡啶组则明显降低外周血CD40,升高CD40L表达(P<0.01),同时降低结肠组织匀浆中IL-1β表达、和升高IL-4表达水平(P<0.05或P<0.01),但灌胃、灌肠组间比较未见明显差异。结论:雷公藤多苷可能通过调节CD40/CD40L表达水平,抑制促炎因子表达,升高抗炎因子水平而有效缓解溃疡性结肠炎结肠粘膜损伤。Objective: To evaluate effect of Tripterygium Glycosides(TG) on expression of CD40,CD40L in peripheral blood from mice with colitis.Methods: All Kunming mice were divided into five groups: The normal group,the model groups,the enema group treated with 20mg/kg TG,the lavage group treated with 20mg/kg TG,and the induced colotis treated with 60mg/kg salicylazosulfapyridine(SASP).Except for mice from the normal groups,experimental colitis was induced by rectal administration of trinitrobenzene sulfonic acid(TNBS) dissolved in 50% ethanol.The therapeutic effect of TG was evaluated by body weight,wet weight of colon,ratio of colonic weight,pathological change and damage score,and expression of CD40 and CD40L of peripheral blood of mice was tested by flow cytometry.IL-1β and IL-4 in colonic tissue homogenate were analyzed by enzyme linked immunosorbent assay.Result: The wet weight,the ratio of colonic weight and damage score from the treated groups were down-regulated when they were compared with that of the model groups(P0.05 or P0.01).While many relieved phenomenon was observed in the treated groups including the sprinkled damage of or intact mucous membrane,scattered inflammatory cell,fewer desmoplasia and restored ulcer.Compared with the model groups,the level of CD40 was obviously decreased and the CD40L was markedly increased in all of the treated groups(P0.01).While the level of IL-1β in the three treated groups is lower than in the model group.And the IL-4 is higher in all the treated groups than in the model groups(P0.05 or P0.01).But the difference was not seen between the enema and lavage groups with TG.Conclusion: It was realized by regulating expression of CD40/CD40L of peripheral blood to regulate the balance of anti-inflammatory and proinflammatory factor that TG could effectively attenuate damaged colonic mucosa with ulcerative colitis.
