TRAIL联合川东獐牙菜诱导人肝癌细胞的凋亡

摘　　要：目的:探讨重组人TRAIL蛋白联合川东獐牙菜正丁醇部位提取物对HepG2人肝癌细胞的凋亡诱导效应。方法:用MTT法和流式细胞仪检测人重组TRAIL蛋白及川东獐牙菜正丁醇部位提取物单独或者联合作用于HepG2人肝癌细胞后的细胞生长抑制率和凋亡发生率,TUNEL法观察细胞凋亡发生情况。结果:MTT和流式细胞仪检测显示:200μl浓度50μg/ml川东獐牙菜正丁醇部位提取物组肿瘤细胞抑制率为89.48%、凋亡率48.66%。100μl浓度100ng/ml TRAIL联合100μl浓度50μg/ml川东獐牙菜正丁醇部位提取物组抑制率和凋亡率均明显提高,分别为96.98%和65.71%,具有显著性差异。TUNEL显示:TRAIL联合川东獐牙菜正丁醇部位提取物组细胞核棕黄色深染,且阳性染色细胞数目明显增多,表明TRAIL与川东獐牙菜正丁醇部位提取物联合作用凋亡细胞明显增加。结论:TRAIL与川东獐牙菜正丁醇部位提取物具有协同杀灭HepG2人肝癌细胞作用。Objective： To study the apoptosis inducing effect by Tumor necrosis factor related apoptosis-ligand（TRAIL）and Swertia davidi Franch on human hepatic carcinoma cells.Methods： The human hepatic carcinoma cells were treated with TRAIL and Swertia davidi Franch in vitro.The inhibition ratio of tumor cells were determined by MTT colorimetric assay,and the incidence of cells apoptosis were determined by flow cytometry method.The apoptosis inducing effect were detemined by TUNEL.Results： Compared with Swertia davidi Franch group（89.48% and 48.66%）,The inhibition rate of the proliferation and apoptosis rate of Hepatic carcinoma cells in Swertia davidi Franch combined with TRAIL group were higher（96.98% and 65.71%）,by MTT and FCM.Karyopycnosis,nuclear chromosomal condensation and segmentation were observed by TUNEL.Swertia davidi Franch could significantly induce the apoptosis of tumor combined with Swertia davidi Franch.Hepatic carcinoma cells were restrained to apoptosis by TRAIL and Swertia davidi Franch.TRAIL could enhance sensitivity of Swertia davidi Franch to up-regulating the expressions of TRAIL death receptors HepG2 cells.Conclusion： TRAIL and Swertia davidi Franch have killing effects on human hepatic carcinoma cells,which have a promising prospect on the treatment of hepatoma.
