摘  要:目的探讨间歇性低压低氧预处理(Hypobaric Hypoxia Preconditionin,HHP)对大鼠全脑缺血/再灌注(Global cerebral ischemia and reperfusion,GCIR)后海马CA1区神经元迟发性死亡(Delayed Neuron Death,DND)及热休克蛋白70(HSP70)表达的影响。方法将Wistar大鼠48只随机分为四组:假手术组(Sham组)、低压低氧对照组(HHP组)、全脑缺血/再灌注组(GCIR组)、低压低氧预处理+全脑缺血/再灌注组(HHP+GCIR组)。采用吴穹改良的复制间歇性低压低氧环境模型,选用四血管闭塞法复制大鼠GCIR模型。硫堇染色下观察海马CAl区锥体神经元组织学分级(Histological grade,HG)和神经元密度(neuronal density,ND),反映锥体细胞DND程度;免疫组织化学方法观察神经细胞HSP70阳性细胞表达变化,将四组结果进行统计学比较。结果 Wistar大鼠HHP+GCIR组海马CA1区较GCIR组HG分级降低、ND明显增加(P<0.05),HSP70阳性表达明显增高(P<0.05)。结论推测间歇性HHP是通过上调神经细胞HSP70的表达,减少神经元DND,发挥对脑组织的保护作用。Objective To study the effect of intermittent hypobaric hypoxia preconditioning(HHP) on neural DND and expression of HSP70 at Hippocampus CA1 Region of Rat after Global Cerebral Ischemia-reperfusion Injury.Methods The Wistar rats were randomly divided into four groups:Sham group,HHP control group,global cerebral ischemia and reperfusion(GCIR)group and HHP+GCIR group.The hypobaric hypoxia environment model modified by Wuqiong was used and occlusion four vessels method was used to made global cerebral ischemia model.Histological grade(HG)and neuronal density(ND)of hippocampal CA1 pyramidal neurons were observed after stained by thionine to show the degree of DND.Immunohistochemical method was used to detect changes of HSP70 expression.The results of four groups analyzed with SPSS software.Results Compared with GCIR group,HG of hippocampal CA1 in HHP+GCIR group was decreased(P〈0.05)but ND increased(P〈0.05)and expression of HSP70 increased obviously(P〈0.05).Conclusion Intermittent hypobaric hypoxia preconditioning has the effect on protect neural by increasing HSP70 expression and decreasing DND.
