摘  要:目的现场评价虚拟浓缩器采集微生物气溶胶的性能。方法在公共场所现场条件下,使用Biosampler和虚拟浓缩器与Biosampler采样器联用两种采样方法,分别采集军团菌气溶胶以及甲型和乙型流感病毒气溶胶,以巢式PCR扩增嗜肺军团菌Mip基因,一步法逆转录巢式PCR扩增甲型流感病毒基质蛋白基因和乙型流感病毒核蛋白基因,琼脂糖凝胶电泳检测扩增产物。结果共采集军团菌气溶胶样本98份,流感病毒气溶胶样本32份。虚拟浓缩器与Biosampler联用和Biosampler单独使用采集嗜肺军团菌的阳性率分别为12.2%(6/49)和18.4%(9/49),甲型流感病毒的阳性率分别是33.3%(4/12)和25.0%(5/20),乙型流感病毒阳性率分别为41.7%(5/12)和40.0%(8/20)。两种采样方法采集的嗜肺军团菌和流感病毒阳性率差异均无统计学意义。结论虚拟浓缩器可起到浓缩微生物气溶胶的作用,与Biosampler微生物采样器联用可为快速高效地采集低浓度微生物气溶胶提供技术手段。Objective To evaluate the bio sampler with virtual concentrator for sampling low concentration microbial aerosol on site. Methods Legionella aerosols, influenza A virus and influenza B virus aerosols were collected by Biosampler with and without virtual concentrator in public buildings. Nested PCR based on Mip gene was used to detect LegioneUa pneumophila~ One-step RT-nested PCR based on influenza A virus matrix protein gene and influenza B virus nucleoprotein gene were used to detect influenza A and B virus respectively. PCR products were detected by agarose gel electrophoresi. Results A total of 98 Legionella and 32 influenza virus aerosol samples were collected. The positive rate of LegioneUa, influenza A virus and influenza B virus which collected by Biosampler with concentrator and without were 12.3%(6/49) and 18.4%(9/49), 33.3%(4/12) and 25.0%(5/20), 41.7%(5/12) and 40%(8/20) respectively. There was no significant difference in the positive rate of Legionella aerosol, influenza A virus and influenza B virus aerosol between the two sampling methods (P=0.453 ;P= 0.696;P=1.000). Conclusion The virtual concentrator combined with biosampler can be used to quickly sample the low concentration microbial aerosol on site.
