摘 要:目的探讨宣威肺癌高发区室内灰尘对人体肺腺癌细胞株A549和人脐静脉血管内皮细胞株EA.hy 926细胞存活率的影响。方法 A549和EA.hy 926细胞分别暴露于终浓度为10、25、50、100、200μg/ml室内灰尘溶液,PBS为溶剂对照,24h后,采用MTS法测定细胞存活率。结果室内灰尘均能抑制A549和EA.hy926细胞增殖(P<0.05),增加细胞死亡(主要为坏死),有剂量-反应关系。在相同染毒浓度下A549的存活率低于EA.hy926的存活率。结论体外实验发现宣威室内灰尘对A549和EA.hy926细胞均有细胞毒性,但A549细胞对宣威室内灰尘更为敏感。Objective To investigate the effect of the indoor dust from lung cancer high-risk areas in Xuanwei, Yunnan, China on the cellular viability of human umbilical vein endothelial cell EA. hy926 and human lung adenocarcinoma cancer cell A549. Methods A549 and EA.hy926 cells were exposed to indoor dust at the doses of 10, 25, 50, 100 and 200 μg/ml respectively, PBS solvent was taken as the control. After 24 hours of treatment, the cellular viability was measured by using the MTS method. Results All exposure doses of the indoor dust can reduce the proliferation of EA. hy926 and A549 cell (P〈 0.05), increase cell death (mainly necrosis) with a dose-dependent manner. In the same exposure concentrations, the cellular viability of A549 was lower than the survival rate of EA.hy926. Conclusion A549 cells are more sensitive to indoor dust from Xuanwei compared with EA.hy926 cells.
