解毒清肺合剂对慢性阻塞性肺疾病急性加重期痰热蕴肺证模型大鼠气道杯状细胞的影响

摘　　要：目的观察解毒清肺合剂对慢性阻塞性肺疾病急性加重期痰热蕴肺证模型大鼠气道黏液高分泌的影响。方法 96只大鼠随机分为对照组、模型组、中药治疗组,模型组和中药治疗组采用香烟烟雾暴露与脂多糖气道内滴入联合造模方法制备模型大鼠。实验第1日起,中药治疗组以解毒清肺合剂按1 mL/100 g剂量灌胃,每日1次;模型组以生理盐水按1 mL/100 g剂量灌胃,每日1次;空白对照组正常喂养。于实验第10、20、30、40日分期取材,取肺组织行阿尔辛蓝-过碘雪夫染色,计算气道杯状细胞数目。结果与对照组比较,模型组各期杯状细胞数目均明显增多,差异有统计学意义(P<0.05);与模型组比较,中药治疗组第20、30、40日时杯状细胞数目明显减少,差异有统计学意义(P<0.05)。结论解毒清肺合剂可以减少慢性阻塞性肺疾病急性加重期痰热蕴肺证模型大鼠气道杯状细胞的化生,减少气道黏液高分泌。Objective To research the effect of Jiedu Qingfei mixture on airway mucus hypersecretion of rats with acute exacerbation of chronic obstructive pulmonary disease（AECOPD）.Methods A total of 96 rats were randomly divided into control group,model group and TCM treatment group.Rats in TCM treatment group and model group were treated by cigarette smoking and LPS dripping to make model,and were given Jiedu Qingfei mixture and saline in dosage of 1 mL/100 g per day respectively.The rats in each group were killed at 10th,20th,30th,40th day respectively,and were performed with AB-PAS stain to count goblet cell.Results There was significant difference between control group and model group in each stage at 20th,30th,40th day（P〈0.05）.There was a significant difference between treatment group and model group（P〈0.05）.Conclusion Jiedu Qingfei mixture can reduce airway mucus hypersecretion of rat model with AECOPD by relieving goblet cell metaplasia
