摘 要:目的观察吡咯喹啉醌(PQQ)对兔骨关节炎模型关节软骨的保护作用。方法Huhh法建立日本大耳白兔右侧膝关节骨关节炎模型;实验组兔术后给予关节腔内注射1×10^4μmol/LPQQ,0.2ml/kg,3d1次,连续6周;对照组兔行关节腔内等量生理盐水注射;伪手术组兔仅暴露关节,不进行药物处理。术后6周行膝关节前后位X线检查,并行Kellgren-Lawrance评分;观察股骨髁关节面大体形态并行软骨PelletierJP评分;制备切片行苏木素一伊红(HE)染色,光镜观察、Mankin评分;行Ⅱ型胶原免疫组织化学染色;原位末端转移酶标记(TUNEL)法检测软骨细胞凋亡;透射电镜下观察软骨细胞线粒体形态和数目的改变。结果实验组、对照组和伪手术组Kellgren—Lawrance评分为1.49±0.45、5.69±1.08、0.28±0.06;JP评分为1.46±0.42、2.75±0.60、0.08±0.04;膝关节组织学Mankin评分为2.68±0.86、6.95±1.46、0.25±0.06;凋亡软骨细胞所占总细胞数目的比例为(5.60±1.20)%、(35.30±4.90)%、(0.704-0.30)%;实验组、对照组和伪手术组电镜下软骨细胞线粒体数为3.40±0.68、1.24±0.45、6.40±1.20;上述各指标组间比较差异有统计学意义(P〈0.05)。实验组免疫组织化学染色可见Ⅱ型胶原纤维组织结构良好,而对照组软骨Ⅱ型胶原缺失。结论关节腔内注射PQQ可保护软骨细胞,减轻关节软骨结构及功能的破坏,减缓关节软骨退变。PQQ可能是通过保护软骨细胞线粒体而发挥作用。Objective To study the protective effect of pyrroloquinoline quinone (PQQ) on carti- late in rabbit osteoarthritis models. Methods Thirty-six 6-month-old Japanese white rabbits were randomly divided into experimental group, control group and sham-operation group (12 rabbits in each group). The osteoarthritis models at the fight knee in experimental group and control group were established followed by the Huhh method. Animals in the experimental group were given intra-artieular injection of 1 × 104 μmo]/L PQQ (0. 2 ml/kg body weight), 3 days a week for 6 weeks. The same volume of saline was delivered in the control group. There was no special treatment on animals in the sham-operation group. Animals were sacrificed 6 weeks after anteroposterior X-ray evaluation. HE staining, type II collagen immunohistoehemi- eal staining, chondroeyte apoptosis [ TdT-mediated dUTP nick end labeling (TUNEL)] detection, and e- lectron microscopy observation of mitochondrion changes in cartilage were done. Results Kellgren-Law- rance score in experimental group, control group and sham-operation group was 1.49 ± 0. 45, 5.69 ± 1.08, and 0. 28 ± 0.06, respectively,P 〈 0.05. Degeneration changes of femoral condyle were alleviated by PQQ ( JP score 1.46 ± 0. 42, 2.75 ± 0. 60, 0. 08 ± 0. 04, P 〈 0.05 ). Mankin scores were 2.68 ± 0. 86, 6. 95 ± 1.46 and 0. 25 ±0. 06,P 〈0. 05 ; and number of apoptotic chondrocytes was (5.60 ± 1.20)%, (35.30 ± 4. 90) %, (0.70 ± 0. 30) % , P 〈 0. 05. More mitochondrials were observed under an electron microscope (3.40 ± 0. 68, 1.24 ± 0.45, and 6.40 ± 1.20, P 〈 0. 05 ). Well structured type II collagen fibers were observed in the experimental group. Conclusion Intra-articular injection of PQQ can protect the cartilage during osteoarthritis development by alleviating the structure and function damage of the cartilage and pro- moting its metabolism and self-repair capacity. PQQ may take the function by protecting mitochondria in chondrocytes.
