摘 要:目的探讨螺内酯(spironolactone,SPI)对高糖诱导的大鼠肾小球系膜细胞(glomerular mesangial cells,GMCs)的增殖和核因子-κB(nuclear factor-kappa B,NF-κB)表达及活性的影响。方法建立高糖培养条件下的大鼠GMCs模型,采用噻唑蓝法(MTT)检测各组(根据葡萄糖及螺内酯浓度来分组)大鼠GMCs增殖情况,半定量(reverse transcriptase-polymerase chain reaction,RT-PCR)法及细胞免疫荧光检测不同条件下的NF-κB的表达和活性。结果高糖培养组大鼠GMCs增殖明显高于低糖培养组(P〈0.05)。加入螺内酯后,GMCs增殖受到抑制,与低糖、高糖组比较(P〈0.05),且随着螺内酯浓度增加细胞增殖抑制递增。螺内酯可下调高糖诱导的NF-κB(messengerRNA,mRNA)的表达和活性,具有浓度依赖性。结论高糖增强大鼠肾小球系膜细胞的增生作用通过NF-KB参与调控。螺内酯可在基因水平抑制高糖刺激的大鼠GMCs的NF-κB表达与活性。Objective To study the effects of spironolactone on cell multiplication and expres- sion of NF-κB and the activity of NF-κB. Methods The model of rat mesangial ceil was set up in high glucose. The NF-κB expression at mRNA and and the activity of NF-κB at cell levels under different conditions was determined by RT-PCR and immunofluorescence. Results GMCs proliferation in high glucose is high than that in low glucose(P〈0. 05). The GMCs proliferation was inhibited when the different concentrations of spironolactone were added(P〈0. 05). Spironolactone strongly inhibited cell proliferation in a dose-and- time-dependent manner. Spironolactone could decrease the levels of NF-κB mRNA and the activity of NF-κB. Its effect was in a dose-depended manne. Conclusions High glucose could enhancs the hyperplasia of rat GMCs and the effect was dependent on NF-κB.
