摘 要:目的研究碳纤维复合材料粉尘(CFC)的细胞毒性作用。方法以小鼠肺成纤维细胞(NIH/3T3)为靶细胞,以某飞机制造企业使用的碳纤维复合材料沉降尘为受试物,采用噻唑蓝(MTT)比色法检测碳纤维复合材料粉尘对NIH/3T3细胞活性与增殖的影响,并计算相对增殖率(RGR);采用单细胞凝胶电泳(SCGE)技术检测NIH/3T3细胞的DNA损伤情况。结果经受试物染毒的细胞数目减少,有死亡细胞,随着染毒浓度的增加,存活的细胞数逐渐减少,并出现细胞吸附毒物的现象。不同浓度下的碳纤维复合材料粉尘(100、200、400、800μg/ml)作用一定时间后,NIH/3T3细胞存活率下降,呈现出时间-剂量-反应关系。碳纤维复合材料粉尘可引起NIH/3T3细胞DNA断裂,染毒组的细胞拖尾率、尾部DNA含量、彗星细胞的Olive尾距、延伸的尾动差均增高,与阴性对照组比较,差异有统计学意义(P<0.05)。结论碳纤维复合材料粉尘具有明显的细胞毒性,并对DNA有损伤作用。Objective To evaluate the cytotoxicity of carbon fiber composite dust. Methods The NIH/ 3T3 cells of mice were taken as the target cell and settled dust of carbon fiber composite from aircraft manufacturing workshop as the experiment material. The viability and proliferation of NIH/3T3 cells were tested by MTT assay. The DNA damage was determined with single ceil gel electrophoresis (SCGE) technique. Results After exposure the number of cells reduced~ dead cells were found. As the exposure dose increased, viable cells gradually decreased, and the phenomenon of cell with adsorbed poison appeared. After reacted with different concentrations of carbon composite dust(100,200,400, and 800 μg/ml)for a certain time, the viability of NIH/3T3 cells reduced showing a time-dose-response relationship. Carbon fiber composite dust could also break the cell DNA into fragments. In exposed group all the percentages of tailed cells, olive tail moment and extent TM increased, the difference was significant in comparison with those of the control group(P〈0. 05). Conclusions The carbon fiber composite dust has evident cytotoxicity and could damage the DNA.
