摘  要:目的探讨焦炉逸散物对焦炉工人端粒长度和端粒酶活力变化的影响。方法以焦炉作业工人215名为接触组,非焦炉作业工人125名为对照组。采空腹外周静脉血,提取外周血中白细胞基因组DNA和端粒酶,应用实时荧光定量聚合酶链式反应(PCR)检测外周血中白细胞端粒DNA相对长度、端粒酶相对活力。结果接触组外周血中白细胞端粒DNA相对长度小于对照组,端粒酶相对活力高于对照组,差异均有统计学意义(P<0.05)。接触组端粒相对长度与端粒酶活力呈明显负相关(r=-0.506,P<0.01)。炉顶工、炉底工组、炉侧工组的端粒相对长度、端粒酶活力与炉顶、炉底、炉侧的苯并芘B(a)p浓度分别呈明显负相关(r=-0.472,P<0.01)和正相关(r=0.421,P<0.01)。结论焦炉逸散物可导致端粒缩短、端粒酶活力增高,从而引起端粒稳态失衡。Objective To explore the impact of coke oven emission on the changes of telomere length and telomerase activity in coke oven workers. Methods 215 coke oven workers were taken as exposed group. 125 employees who had no exposure history to coke oven emission were selected as the control group. Use the test kits to extract leukocyte genuine DNA and telomerase from their peripheral venous blood. The SYBR Green I real time PCR were used to detect the relative telomere length and relative telomerase activity. Results Relative telomere DNA length of white blood cells in the exposure group was shorter than that of the control group and relative telomerase activity in the exposure group was stronger than that of the control group. All the differences were statistically significant(P〈0.05). The relations of relative telomere DNA length and relative telomerase activity in the exposure group were a significantly negative correlation(r= 0. 506, P〈 0. 01). The relations of relative telomere DNA length in the workers working on oven top, oven bottom and oven side and the concentration of B(a)p at the oven top, oven bottom and oven side were significantly negative correlation(r=- 0. 472 ,P〈0.01), but the relations of relative telomerase activity showed positive correlation (r=0. 421, P〈0.01). Conclusions Coke oven emission could shorten the telomere DNA, and elevate the activity of telomerase, causing the disruption of telomere homeostasis.
