摘 要:采用RAPD分子标记技术分析9个自然居群米心水青冈的遗传关系和多样性。20条随机引物共扩出260个条带,平均每条引物扩增出13个条带,其DNA带的分子量在200~3000 bp。其中多态性条带有180条,多态位点比率(PPL)为69.2%。种内Shannon多样性指数为0.302 1,群体内所占比例为72.10%,群体间所占比例为27.90%。种内Nei's基因多样性为0.204 5,群体内平均基因多样性为0.150 2,基因分化系数GST为0.263 1。米心水青冈的遗传多样性水平处于中等,遗传分化也处于中等水平。The genetic relationship and genetic diversity of nine natural populations of Fagus engleriana from four provinces were analyzed by random amplified polymorphic DNA (RAPD) marker technique. Totally 260 bands were amplified by 20 primers, 13 bands were averagely amplified by each primer. The molecular weight of the band DNA was between 200 bp and 3 000 bp. Out of the 260 bands, 180 bands were polymorphic and the percentage of polymorphic loci (PPL) was 69.2%. The average Shannon index value within the species of F. engleriana was 0. 302 1, and the PPL within the population and among the nine populations was 72.10% and 27.90% respective- ly. The average Nei' s genetic diversity within the species of F. engleriana was 0. 204 5, and 0. 150 2 for the index within the population. The coefficient of genetic differentiation was 0. 263 1. Compared with other species, the ge- netic diversity of F. engleriana was at a moderate level. And its genetic variance level within the populations was also at a moderate level.
