摘 要:目的:评价复方中药"天芪航力片"对大鼠正加速度(+Gz)致心肾损伤的防护效果,为减轻高G损伤提出新的防护措施。方法:将36只雄性SD大鼠随机分为3组(每组12只),即对照组(A组)、+Gz组(B组)和实验药物组(C组)。C组连续给予复方中药"天芪航力片"提取物14 d,每天1次,给药量为20 ml/kg。A组和B组每天给予等量的蒸馏水。14 d后,B组和C组进行+10 Gz持续5 min,增长率为1 G/s的高G暴露。各组均于+Gz作用后1 h,采集血样分离血清,送第四军医大学西京医院临床免疫科测定大鼠血清心脏损伤标志物肌酸激酶(CK)和肌酸激酶同工酶(CK-MB),以及肾脏损伤标志物血清尿素氮(BUN)和肌酐(Cr)的含量。对肾脏ATP酶的表达进行组织化学染色法检测。取肾脏组织超薄切片进行电镜观察。结果:B组血清CK和CK-MB以及C组的CK-MB含量较A组均显著升高(P<0.01),而C组CK的含量较A组显著升高(P<0.05)。B组大鼠血清BUN的含量较A组显著降低(P<0.05),血清Cr的含量显著升高(P<0.05)。C组血清BUN及Cr的含量与A组比较无显著变化,但与B组相比BUN含量有显著性差异(P<0.05)。B组肾脏ATP酶阳性细胞数较A组显著降低,而C组ATP酶阳性细胞数则无明显变化。B组肾脏超微结构出现明显损伤性改变而C组则无明显变化。结论:复方中药"天芪航力片"提取物对+Gz引起的心肾损伤具有一定的防护作用。AIM: To evaluate the protective effect of the extraction of Fufang Tianqi tablet, a compound prescription of Chinese medicinal herbs, on cardiac and kidney injuries caused by + Gz in rats and to propose new protective measures for high + Gz. METHODS: Thirty-six male Sprague Dawley rats were randomly divided into three groups : control group ( group A), high + Gz group ( group B) and drug group (group C). The rats in group C were treated with Fufang Tianqi tablet extraction for 14 days, whereas rats in group A and group B were treated with the same amount of water (20 ml/kg). Rats in group B and group C were exposed to + 10 Gz for 5 min after 14 days. At 1 h after centrifugation, blood samples were taken from rats in all three groups for the assaying of heart and kidney injury markers con- tent in the Department of Clinical Immunology, Xijing Hospital. The kidney tissues were taken for kidney ATP enzyme staining and electron microscopy observation. RESULTS: Compared with those in group A, CK and CK-MB significantly increased in group B. CK-MB also significantly increased in group C after high + Gz exposure (P 〈0. 01 ). In addition, CK significantly increased in group C with group A (P 〈 0.05). Compared with those in group A, BUN reduced significantly in group B ( P 〈 0. 05 ), and Cr significantly increased (P 〈 0. 05 ). BUN and Cr showed no significant change in group C with group A, but BUN significantly increased in group C compared with group B ( P 〈 0. 05 ). Renal ATP content sig- nificantly reduced in group B compared with that in group A after + Gz exposure. Electron microscopy showed that kidney damage in group B was more significant than in group C. CONCLUSION: Fufang Tianqi tablet extraction exerts some protective effect on the improvement of ischemia and hypoxia in rats after high + Gz exposure.
