摘 要:目的观察加味补中益气汤对大鼠变应性鼻炎(AR)晚发相脾组织IL-5表达水平的影响。方法制备AR大鼠,随机分为模型组、加味补中益气汤中药组和地塞米松西药组,另设正常大鼠组。模型成功后用药10 d,末次灌胃并鼻腔激发后,按30 min~1 h,1 h~2 h,2 h~4 h,4 h~6 h,6 h~8 h,8 h以后共6个时间段分批取鼻黏膜和脾组织,HE染色计数其嗜酸性粒细胞(EOS)数,ELSIA法检测脾IL-5水平。结果加味补中益气汤能有效降低IL-5的表达和EOS数,在第3、4时间段之后即有显著差异(P<0.01),且第5、6时间段中药组的IL-5含量和EOS数明显低于西药组(P<0.01)。结论加味补中益气汤能降低IL-5表达,减少鼻黏膜嗜酸性粒细胞的浸润。Objective To investigate the inhibitory effects of Modified Center-Supplementing Qi-boosting Decoction(MCSQBD) on the expression of IL-5 in spleen tissue of rats with experimental allergic rhinitis in the pathological process.MethodsThe rat model of allergic rhinitis was established at first based on a routine procedure.Then,the successfully modeled animals were randomly divided into 3 groups,i.e.modeling group(MG) treated with only normal saline,treating group(TG) treated with MCSQBD and positive controlling group(PCG) treated with dexameshasone,with 27 rats in each group.Moreover,another 26 healthy rats were taken as normal controlling group(NCG).Once the treating course lasted for 10 days ended,all the animals were put into death in a regular 6 time intervals as 30 min^1 h,1 h^2 h,2 h^4 h,4 h^6 h,6 h^8 h and 8 h to take samples of serum,spleen and nasal mucosa respectively with the samples stored routinely.The expression of IL-5 was determined by enzyme linked immunosorbent assay(ELISA),and the infiltrating density of eosinophils(EOS) in nasal mucosa was detected by pathohistological procedures,with the pathological changes in nasal epithelia observed meantime for a comparison among all groups.Results The results from this study indicated that MCSQBD could significantly inhibit the expression of IL-5 in spleen,especially during the time interval Ⅲ after interval Ⅱ,i.e.the late phase of AR.This kind of effect of MCSQBD was shown significantly superior to that of dexameshasone(p=0.003).Conclusion MCSQBD can effectively inhibit the expression of IL-5 in lymphatic system of rats with experimental allergic rhinitis,especially at its late phase,with a paralleled decreasing of EOS infiltrating density in nasal mucosa.Then,allergic reaction in nasal mucosa should be reduced in this way.
