不同批号葛根芩连微丸中葛根素、黄芩苷和盐酸小檗碱的同步测定及含量比较

摘　　要：目的建立HPLC同时测定葛根芩连微丸中葛根素、黄芩苷、盐酸小檗碱3种有效成分的含量的方法,并比较三个批次葛根芩连微丸此三种主要活性成分的含量。方法色谱柱为铂金ODS(250 mm×4.6 mm,5μm)柱,流动相为乙腈-水(含0.025%磷酸+0.035%三乙胺),梯度洗脱,流速1 ml.min-1,检测波长270 nm,柱温30℃。结果葛根素在0.0358～1.430 8μg范围内与峰面积具有良好的线性关系(r=1),平均回收率为101.2%,RSD=0.60%(n=6);黄芩苷在0.0191～0.7631μg范围内与峰面积/10 000具有良好的线性关系(r=1),平均回收率为100.36%,RSD=1.38%(n=6);盐酸小檗碱在0.049 9～1.998 0μg范围内与峰面积/10 000具有良好的线性关系(r=1),平均回收率为102.16%,RSD=0.36%(n=6);三批微丸中葛根素,黄芩苷和盐酸小檗碱含量的均值分别为16.11mg/g,11.01 mg/g和17.22 mg/g。结论方法准确、灵敏、重现性好,阴性无干扰,可用于葛根芩连微丸中3种有效成分的同步测定;葛根芩连微丸3批之间3种成分含量差异显著。Objective To establish an HPLC method for simultaneous qualitative and quantitative determination of puerarin,baicalin and berberine hyhrochloride in Gegenqinlian pill and Gegenqinlian Decoction and to compare the content of three bioactive components between pill and decoction of Gegenqinlian. Methods PLATISILTM-ODS（250mm × 4.6mm,5μm） column was used at 30℃,with mobile phase consisted of acetonitrile-water（containing 0.025% phosphoric acid ＋0.035% triethylamine）（gradient elution） at a flow rate of 1ml·min-1,and 270nm as detected wavelength. Results The puerarin showed good linearity in the range of 0.0358~1.4308μg（r = 1.0000） and the average recovery was 101.2%,RSD =0.60%（n=6）.The baicalin showed good linearity in the range of 0.0191 ~0.763 1 μg（r = 1.0000）and the average recovery was 100.36%,RSD = 1.38%（n=6）.The berberine hydrochloride showed good linearity in the range of 0.0499-1.9980μg（r = 1.0000） and the average recovery was 102.16%,RSD = 0.36%（n=6）.Three batches of pills of puerarin,baicalin and berberine hydrochloride were respectively a value of 16.11mg / g,11.01 mg / g and 17.22 mg / g. Conclusion The method is accurate,sensitive,reproducible and is suitable for the quality control of three bioative components in Gegen Qinlian pill and Gegen Qinlian Decoction.The three bioative components in the three batches Gegen Qinlian pill have significantly different centents.
