摘 要:目的探讨银杏黄酮对CCl4诱导的小鼠肝损伤的保护作用及其机理。方法 60只ICR小鼠随机分为5组,即空白对照组、模型组和银杏黄酮低、中、高剂量组,采用CCl4腹腔注射和灌胃制作小鼠肝损伤模型。通过检测血液中谷丙转氨酶(ALT)和谷草转氨酶(AST)活性、观察肝脏的解剖学形态和病理切片评估其对小鼠肝脏的保护作用。RT-PCR检测PXR、CYP3A11、CYP3A13和RXRα的表达。结果与空白对照组相比,模型组小鼠肝脏出现肿胀、色泽暗淡、轮廓萎缩、表面粗糙;血清中ALT和AST活性明显升高。与模型组相比,银杏黄酮组小鼠肝脏肿胀消退、色泽红润、轮廓清晰、表面光滑;血清中ALT和AST活性明显降低,肝组织中PXR、CYP3A11、CYP3A13和RXRα水平显著升高。结论银杏黄酮对CCl4所致的小鼠肝损伤具有一定的保护作用,且其保护作用呈明显的剂量依赖性。PXR、CYP3A11、CYP3A13和RXRα等基因的激活可能在保护小鼠肝损伤中起到重要作用。Objective To investigate the protective effects of ginkgetin against carbon tetrachloride(CCl4)-induced hepatic injury in mice and discuss its mechanism.Methods60 ICR mice were randomly divided into five groups:control,model and small,middle and large dosage of ginkgetin groups.The model of hepatic injury was established through intragastric administration and injection with CCl4.The protective effects of ginkgetin against hepatic injury in mice were evaluated by detecting the activities of serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) and observing morphology and pathological section of liver.PXR,CYP3A11,CYP3A13 and RXRα were detected by RT-PCR.Results Compared with the control,livers of model group appeared swelling,dark red,shrank contour and rough surface,and the activity of serum ALT and AST increased significantly.While compared with model group,livers of ginkgetin groups appeared no swelling,normal color,integral contour and smooth surface,the activity of serum ALT and AST decreased significantly,and levels of PXR,CYP3A11,CYP3A13 and RXRα in liver increased significantly.Conclusion Ginkgetin can prevent liver from injury induced by CCl4 to some extent,and the protective effects are dose-dependant remarkably.Activation of PXR,CYP3A11,CYP3A13 and RXRα gene may play an important role in protection of hepatic injury in mice.
