摘  要:目的通过构建巨噬细胞炎症模型,研究中药单体野黄芩苷对MAPK信号通路及细胞因子基因表达的影响。方法培养巨噬细胞RAW264.7,利用脂多糖构建细胞炎症模型,并在不同时间收集细胞。Western blot与RT-PCR法分别检测野黄芩苷对MAPK信号通路及细胞因子基因表达的影响。结果野黄芩苷能够显著抑制细胞炎症模型中JNK/SAPK与p38 MAPK信号通路的磷酸化水平,并且能够降低炎症因子TNF-α的基因表达水平,及提高抗炎细胞因子IL-10、TGF-β的基因表达水平来发挥其抗炎作用。结论野黄芩苷主要是通过抑制JNK/SAPK与p38 MAPK信号通路的磷酸化水平及提高抗炎细胞因子的基因表达水平来发挥其抗炎作用。Objective Through the construction of inflammation model we observed effects of scutellarin on MAPK signal pathway and gene expression of cytokines. Methods Inflammation model was induced with LPS through cell culture with macrophage RAW264.7 and cells were collected at different times.MAPK signal pathway and gene expression of cytokines were detected with methods of Western Blot and RT-PCR. Results Scutellarin effectively inhibited the phosphorylation of JNK/SAPK and p38 MAPK signal pathway and gene expression of cytokine TNF-α on inflammation model.Scutellarin also exerted anti-inflammatory effects through enhancement of gene expression of cytokine IL-10 and TGF-β. Conclusion It showed that scutellarin exerted anti-inflammatory effects through inhibition of phosphorylation of JNK/SAPK and p38 MAPK signal pathway and increase of gene expression of anti-inflammatory cytokines IL-10 and TGF-β.
