摘 要:目的探讨大鼠脑缺血再灌注损伤(MCAO)后神经干细胞的增殖分化相关细胞因子IGF-1 mRNA的变化情况和通心络对其影响。方法采用大鼠缺血再灌注损伤(MCAO)模型,采用逆转录集合酶链式反应(RT-PCR)检测了缺血再灌注损伤后不同时间神经干细胞增殖分化相关细胞因子IGF-1 mRNA的变化。结果 IGF-1 mRNA在造模后第3天开始表达增强,随缺血再灌注时间的延长,PCR表达增加,其中造模后第7天表达最高,造模后第14天IGF-1 mRNA表达下降,第30天恢复到基线水平。通心络治疗后第5,7,14,21,30天时IGF-1 mRNA表达均高于缺血对照组。结论大鼠缺血再灌注损伤后可引起其缺血侧神经干细胞增殖分化相关细胞因子IGF-1 mRNA的变化;通心络可显著增强缺血再灌注损伤后大鼠神经干细胞分化相关细胞因子IGF-1 mRNA的表达。Objective To investigate the activation of IGF-1 mRNA after middle cerebral artery occlusion in rats and treatment with Tongxinluo. Methods Ischemia was induced by temporary middle cerebral artery occlusion(MCAO).During the studies of Tongxinluo,the experiment,with RT-PCR,has tested the changes of IGF-1 mRNA expression in different periods. Results The expression of IGF-1 mRNA became more obvious after 3 days.With continuous ischemia and reperfusion,PCR expression increased,and on the 7th day after MCAO,expression was the highest.On the 14th day,PCR expression decreased.It dropped to the baseline level on the 30th day.On the 5th,7th,14th,21 st,and 30th days,PCR expression in groups with the treatment of Tongxinluo was higher than MCAO group. Conclusion Focal cerebral ischemia in the rat results in changes of IGF-1 mRNA expression,and Tongxinluo may enhance the expression of IGF-1 mRNA in the neural stem cells after MCAO.
