摘  要:目的观察脂肪源性干细胞(adipose-derived stem cells,ADSC)体外成骨诱导后的免疫原性。方法体外原代培养ADSC,成骨诱导1、7、14、21 d后,采用流式细胞仪检测各组主要组织相容性复合体Ⅱ类(MHC-Ⅱ)的表达,单向混合淋巴细胞培养(mixed lymphocyte culture,MLC)测定刺激指数(stimulation index,SI)。结果 ADSC成骨诱导后1、7、14、21 d均未检测到MHC-Ⅱ表达;MLC未测到细胞明显增殖(1.15±0.12~1.16±0.18,P>0.05)。结论 ADSC体外成骨诱导后免疫原性低表达,适合作为异体移植细胞。Objective To study the the immunogenicity of ADSCs after differentiation into osteocytes. Methods ADSCs isolated from rat were cultured and induced into differentiate into osteocytes. Before induction and on day 1, 7, 14 and 21 after induction, MHC- Ⅱ was detected with flow cytometer (FCM), stimulation index was examined by One-way MLC. Results ADSCs after osteocytes induction did not express MHC- Ⅱ , and there were no significant changes of stimulation index (from 1.15 ± 0.12 to 1.16 ± 0.18,P〉0.05). Conclusion The immunogenicity of MSCs after differentiation into bone is low, and ADSC is suitable as seed cells of allograft.
