摘  要:目的将培养扩增的兔膀胱移行上皮细胞接种到自制的兔膀胱脱细胞基质(BAM)上构建组织工程复合物,将其种植于皮下观察上皮细胞及BAM的生长分化情况。方法取兔膀胱制备BAM片,将原代兔膀胱移行上皮细胞进行体外培养、扩增后接种到BAM片上体外培养7天后种植到皮下,分别于2周和4周取材行HE及免疫组化检测。结果2周后取材发现复合物挛缩不明显,4周后取材发现复合物挛缩明显,瘢痕形成,条带状上皮细胞生长不明显。结论瘢痕挛缩可能是组织供血不足造成的,移植物血管化成为组织工程化组织应用到体内必须解决的问题。Objectives Tissue - engineered complex using bladder epithelial cells seeded bladder acellular matrix( BAM), was grafted to the subcutis of the rabbit to observe the growth of BAM and epithelial cells. Methods BAM of rabbits were made in vitro. Bladder epithelial cells of rabbits were cultivated and expanded in vitro, then seeded to BAM and cultured for 7 days. The complex was transplanted to the subcutis of the rabbit. Specimens were obtained at 2 weeks and 4 weeks respectively after surgery and were detected by HE and immunohistochemistry. Re- sults No obvious contracture was found at 2 weeks while obvious contracture and scar was found at 4 weeks. Conclusions The reason of the eontraeture may be the lack of blood supply of the complex. The vascularization of graft should be resolved before being applied in vivo.
