摘 要:目的探讨葛花总黄酮(TFF)对糖尿病小鼠视网膜抗氧化损伤的作用。方法 40只C57BL/6J小鼠随机分为5组:正常对照组、糖尿病(DM)模型组、TFF小剂量组(TFFⅠ)、中剂量组(TFFⅡ)和大剂量组(TFFⅢ)。DM、TFFⅠ、TFFⅡ和TFFⅢ组造模成功后第5周开始给药,给药10周,于第15周测定小鼠血糖和体重,处死小鼠取眼球,测定各组小鼠视网膜过氧化终产物丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果与模型组(血糖20.98±5.57 mmol/L,体重21.55±1.67g)比较,给予TFF中剂量组血糖(13.51±6.09 mmol/L)和大剂量组血糖(9.66±2.86 mmol/L)显著下降(P<0.01),大剂量组(体重24.50±1.58g)可明显改善体重的下降(P<0.01)。与模型组(MDA含量:12.10±1.06nmol/mgprot,SOD活性:8.45±0.95 U/mgprot)相比,TFF小、中、大剂量组MDA含量均明显降低(MDA含量分别为:7.98±0.40、5.75±0.63、5.24±0.48 nmol/mgprot,P<0.01),SOD活性均明显升高(SOD活性分别为:13.10±0.90、17.28±0.94、17.79±1.21U/mgprot,P<0.01)。结论 TFF能增强糖尿病小鼠视网膜抗氧化能力,减轻视网膜的氧化损伤,从而对糖尿病视网膜病变起一定保护作用。Objective To explore the antioxidative role of the total flavonoids from Flos Puerariac Lobatae(TFF) in diabetic mice retina. Methods 40 C57BL/6J mice were randomly divided into 5 groups: normal control group ( Con- trol ), diabetic model group ( DM ), TFF low-dose group ( TFF Ⅰ ), medium-dose group ( TFF Ⅱ) and high-dose group ( TFF Ⅲ). The mice were administered with TFF 5 weeks after STZ-induced diabetic model is completed for 10 weeks. And then,the blood glucose and body weight of the mice were measured at the 15 week. Then the animals were submitted to eu- thanasia and the eyeball were collected. The content of end product malondialdehyde (MDA) and activity of superoxide dismutase ( SOD) in mice's retinal were tested. Results Compared with DM group( blood glucose 20.98 ±5.57 mmol/L, body weight 21.55 ± 1.67g) ,the blood glucose of total flavonoids from Flos Puerariae Lobatae medium-dose group( 13.51±6.09 mmol/L) and high-dose group(9.66 ±2.86 mmol/L) was significantly decreased ( P 〈0.01 ), and high-dose group(body weight 24.50 ±1.58g) obviously improved body weight lossing ( P 〈0.01 ). Compared with DM group ( MDA:12.10 ±1.06nmol/mgprot,SOD:8.45 ±0.95 U/mgprot ), the content of MDA of TFF low, medium, and high- dose group was significantly lower ( MDA:7.98±40,5.75 ±0.63,5.24 ±0.48 nmol/mgprot, P 〈0.01 ), and the ac- tivity of SOD of these groups were obviously increased ( SOD : 13.10±0.90,17.28 ±0. 94,17.79± 1.21U/mgprot P 〈 0. 01 ). Conclusion TFF can enhance antioxidative capacity of diabetic mouse retina and reduce the oxidative damage of the retina, which have protective effect on diabetic retinopathy.
