摘  要:目的探讨二甲双胍对人HER2阳性乳腺癌细胞增殖与凋亡的影响。方法不同浓度的二甲双胍干预人HER2阳性乳腺癌细胞SK-BR-3后,采用MTT法检测细胞增殖,流式细胞仪检测细胞凋亡,观察二甲双胍对HER2阳性乳腺癌细胞增殖与凋亡的影响。结果不同浓度的二甲双胍与SK-BR-3乳腺癌细胞共同培养后,随着二甲双胍浓度的逐渐增加,SK-BR-3细胞的增殖抑制也逐渐增加,细胞存活率逐渐下降。统计分析表明,不同浓度的二甲双胍处理组与对照组比较,差异均有显著性(P值均<0.05)。乳腺癌SK-BR-3细胞经二甲双胍处理后,细胞凋亡率(15.09%)明显高于对照组(3.21%),两者比较,差异有显著性(χ2=8.79,P=0.003)。结论二甲双胍对人HER2阳性乳腺癌细胞有增殖抑制及促凋亡作用,其分子机制有待进一步研究。To investigate the effects of metformin on the proliferation and apoptosis of HER2 positive breast cancer cells. [Methods] The HER2 positive breast cancer cells SK-BR-3 were treated with mefformin, growth inhibition ratio of the cells was measured by MTI assay, apoptosis was detected by flow cytometery. IRe- suits ] Breast cancer ceils SK-BR-3 were cultured with mefformin on different concentration, the increased growth inhibition rates of the cells were correlated with the increased concentration of metformin. The growth inhibition ra- tios of metformin (0.5 mM, 2 mM and 8 mM) groups are significantly higher than that of the control group (P 〈0.05). Apoptosis of the mefformin treated group was also significantly different than that of the control group (15.09%, 3.21%, respectively; X2=8.79, P =0.003). [Conclusion] Metformin can inhibit the proliferation and promote the apoptosis in HER2 positive breast cancer cells. Further study is necessary to demonstrate the possible underlying molecular mechanism.
