摘 要:以大白菜品种北京80号、油青矮脚45天菜心、四季菜心王的带柄子叶为外植体,研究不同基因型、苗龄、激素组合、预处理条件及琼脂质量浓度对不定芽再生的影响。结果表明,北京80号的最佳培养基为MS+5mg/L 6-BA+0.5mg/L NAA+2mg/L AgNO3,再生频率为83.6%;油青矮脚45天菜心和四季菜心王的最佳培养基为MS+0.2 mg/L TDZ+1 mg/L NAA+2 mg/L AgNO3,再生频率分别为73.1%和83.2%。3个基因型的带柄子叶在0.5~1mg/L 2,4-D中预处理3d,再生频率达84%以上。4~5d苗龄的外植体诱导不定芽效果较好。琼脂的质量浓度以12g/L为宜。通过不定芽继代培养和生根驯化建立白菜类作物较高再生频率的再生体系。To establish high-frequency shoot regeneration system in Brassica rapa, effects of genotype, seedling age, hormone combination, 2,4-D and agar concentration on shoot regeneration from cotyledonary explants were studied with three varieties including Beijing No. 80, 45 Days Caixin and Four Seasons Caixin. The results showed that the best medium for Beijing No. 80was MS+5 mg/L 6- BA+0.5 mg/L NAA +2 mg/L AgNO3 and the frequency of shoot regeneration could reach 84.1%. The regeneration rate of 45 Days Caixin and Four Seasons Caixin could reach 73.1% and 83.2% respectively when cultured on the medium MS+ 0. 2 mg/L TDZ+1 mg/L NAA+ 2 mg/L AgNO3. Higher frequency was obtained when the explants were pre-cuhured in the MS medium with 0.5-1 mg/L 2,4-D for 3 d before being transferred to the medium with 5 mg/L 6-BA and 0.5 mg/L NAA, the frequency could reach more than 84% The explants from 4-5 days old seedling and the culture medium with 12 g/L Agar are more efficient to induce adventitious buds. A procedure for induction of regenerated shoots and plantlets from cotyledonary explants for Brassica rapa was established in this study.
