摘 要:目的 探讨全反维甲酸在脑损伤早期的保护机制.方法 将55 只SD 雄性大鼠随机分为对照组、盐水对照组,全反式维甲酸组,用Feeney's 法制备脑外伤模型,于8h、1d、2d、3d、7d 五个时间点,测量TGF-β1 mRNA 的表达水平,采用SPSS11.0 软件系统进行统计学分析,P < 0.05 具有统计学意义.结果 在正常大鼠脑组织中有低水平表达,于伤后8h 表达增高;损伤后2~3d 达到高峰,7d 后开始下降.伤后1d、2d、3d、7d 全反式维甲酸组与盐水对照组TGF-β1 mRNA 的表达相比较有显著性差异(P < 0.01).结论 脑外伤后局部及周围组织的TGF-β1 表达水平升高,全反维甲酸能抑制TGF-β1 的表达,对损伤后脑组织的保护与修复有一定的积极作用.Objective To investigate the mechanism of neuron protecting in early injury by all retranic acid and establish the bases for the clinics. Methods 55 SD male fats were derided into control groups, sham groups and all-trans retinoic acid groups (with dose of 10mg/kg/d) , after being injuried by Feeney's test apparatus, groups were operated at 8h, l d, 2d, 3d, 7d devidedly to dected the expression level of TGF-β1 mRNA with inmunocytochemistry and hybridization in situ assay, the IOD of possitive cell were methured, SPSS 11.0 was used in data analysised. Results The expression of TGF-β1 mRNA was low in controls, in sham groups it increased continuously from 8h to 2-3d after injuring, then declined from 7d, the levels at 2-3d and 7d were sharply higher than that at 8h (P〈0.05) ; in aU-trans retinoic acid groups, the levels at ld, 2d, 3d, 7d were severely lower than that in sham groups (P〈0.01) , while the levels at 8h in two groups had no difference (P〉0.05) . Conclusions All-Wans retinoic acid can inhibit the expression of TGF-β1 mRNA it maybe promote neuron repair after being juried.
