红毛五加茎皮抗炎有效成分筛选的初步研究

摘　　要：目的从红毛五加茎皮中提取、分离、筛选抗炎有效成分。方法粉碎红毛五加茎皮,95%乙醇浸渍48h,滤取提取液,合并提取液,减压低温回收溶剂,加水溶解,依次用石油醚、乙酸乙酯、正丁醇萃取。以LPS刺激小鼠巨噬细胞样细胞RAW264.7,加入红毛五加各萃取部分、水相部分共孵育24h,用放射免疫法测定细胞上清液中PGE2的含量,以抑制LPS刺激RAW264.7产生PGE2的作用为指标,逐步对红毛五加提取物进行筛选。结果正丁醇萃取部分的40mg/L以及10mg/L剂量对LPS刺激RAW264.7细胞产生PGE2为(7.26±0.95)pg/mL、(7.43±0.82)pg/mL,与LPS刺激组相比,有统计学意义(P<0.05),抑制率分别为23.7%、21.6%。萃取水相的40mg/L剂量组对LPS刺激RAW264.7细胞产生PGE2为(7.18±1.46)pg/mL,与LPS刺激组相比,有统计学意义(P<0.05),抑制率为24.7%。而其他萃取部分各剂量组与LPS刺激组相比均无统计学意义(P>0.05)。结论红毛五加抗炎的有效成分主要存在于其经乙醇提取,正丁醇萃取部分。Objective To extract, isolat, screen the effective components from Acanthopanax giraldii Harms. stems. Metllods Stems and barks were infused by ethanol （95%） for 48 hours, the liquid was filtered and then was evapned solvent to get extracts. The extracts were dissolved in water, and then extracted by petroleum ether, acetic ether, n-butanol in turn. The n-butanol parts were dried in vacuum and then dissolved in methanol. At last, the n-butanol parts were separated by chloroform-methanol elution system on column with silica. LPS was added to macrophage-like cell RAW264.7 to stimulate PGE2 production. PGE2 production was detected by RIA. Screening of AGH extracts were performed step by step through anti-inflammation index that PGE2 production induced by LPS was inhibited. Results The PGFa production induced by LPS and n-butanol parts （40mg/L, 10rag/L） were （7.26±0.95） pg/mL and （7.43±0.82） pg/mL, respectively. The n-butanol parts inhibited the up-regulation of LPS-induced PGE2 production （vs. LPS; P〈0.05） . Inhibitory ratios of PGE2 were 23.7% and 21.6%, respectively. PGE2 production induced by LPS and water phase parts （40mg/L） was （7.18± 1.46） pg/ mL. The aqueious parts （40mg/L） inhibited the up-regulation ofLPS-induced PGE2 production （vs. LPS; P〈0.05） . ]inhibitory ratio of PGE2 was 24.7%. Conclusion Anti-inflammatory effective ingredient ofAcanthopanax giraldii Harms presented in the n-butanol parts and the aqueious parts.
