摘  要:目的:建立一种准确、快速高效毛细管电泳-紫外检测法测定人血清精氨酸含量的方法。方法:采用高效毛细管电泳法,石英毛细管(70 cm×75μm,有效长度58 cm),分离电压15 kv,检测波长254 nm,高压进样15 s,电泳缓冲溶液比例20 mmol/L pH9.33四硼酸钠:166 mmol/L SDS∶乙腈=10∶10∶3。结果:线性范围为0.03 mmol/L~1.50 mmol/L(r=0.998);以噪声比(S/N)=2时浓度为最低检测限为0.01 mmol/L,精密度为2.4%~4.0%,重复性为2.3%~2.9%,回收率为90.5%~114.2%;检测烧伤患者住院后血中精氨酸含量变化,14 d含量达到高峰。结论:该方法样品预处理简单,分离效果好,可用于实际样品的分析。Objective:To develop a fast and convenient method for determining the content of arginine in human serum.Methods: With high performance capillary electrophoresis,the determination conditions were detection wavelength at 254 nm,fused quartz capillary(70 cm×75 μm,effective length at 58 cm) and a running buffer of 20 mmol/L sodium tetraborate buffer(pH9.33): 166 mmol/L SDS: acetonitrile=10∶ 10∶ 3 at a constant voltage of 15kV and a sampling time of 15 s.Results: The linearity range was 0.03 mmol/L~1.50 mmol/L(r=0.998),the detection limit was 0.01 mmol/L,the precision was 2.4%~4.0%,the repeatability was 2.3%~2.9% and the recoveries of sample were 90.5%~114.2%.The arginine level in normal serum was 0.08 mmol/L,while in serum of burn victims reached its peak in 14 days.Conclusion: The method can be used for the determination of arginine in human serum and the assay results were satisfactory.
