不同稀释液及培养基中氯化钠浓度对炭疽疫苗活菌计数的影响

摘  要:目的研究不同稀释液及培养基中不同氯化钠浓度对炭疽活疫苗活菌数测定的影响。方法分别以氯化钠注射液、磷酸盐缓冲液及注射用水作为稀释液测定3批炭疽疫苗活菌数。将疫苗分别用氯化钠注射液和注射用水作适当稀释,孵育1h后进行活菌数测定。将疫苗以注射用水稀释后接种含不同氯化钠浓度的厚氏琼脂培养基,测定炭疽疫苗活菌数。结果保存3个月疫苗以注射用水作稀释液的活菌数结果比氯化钠注射液高67%,比磷酸盐缓冲液高53%;保存12个月疫苗以注射用水作稀释液的活菌数结果比氯化钠注射液高63%。经氯化钠注射液孵育后的活菌数结果较注射用水孵育后明显降低。以注射用水稀释后菌液接种含不同氯化钠浓度的琼脂培养基,活菌数无明显差异。结论注射用水是炭疽活疫苗活菌数测定的合适稀释液,培养基中氯化钠对活菌数测定无明显影响。Objective To study the effects of different dilution solution and different concentration of sodium chloride in medium on viable count of anthrax vaccine. Methods Anthrax vaccine were diluted with three kinds of diluents, including sodium chloride injection, PBS and water for injection, the viable counts were measured and compared. The viable count of anthrax vaccine after incubated in sodium chloride injection or water for injection were measured. The viable count of anthrax vaccine with medium containing different concentration of sodium Chloride were determined. Results The viable count of anthrax vaccine diluted with water for injection was much higher than that of sodium chloride injection and PBS. The measured result of vaccine after incubated in sodium chloride injection was lower than that of water for injection. There was no statistical difference of viable counts comparing medium with different sodium chloride concentrations. Conclusion Using water for injection as diluent is suitable for assay of viable count of anthrax vaccine. The concentration of sodium chloride in medium did not affect the measured results.
