摘  要:目的:探讨美洲大蠊提取物(CⅡ-3)对荷瘤小鼠S180肉瘤的抑制作用。方法:昆明种小鼠右前肢腋下接种S180细胞,接种次日随机分为模型组、环磷酰胺组(30 mg·kg-1,隔天ip)、CⅡ-3高、中、低剂量组(200,100,50 mg·kg-1,ig),连续给药10 d,同期设正常组;观察CⅡ-3对S180肉瘤生长的作用,称量小鼠免疫器官质量,计算脏器指数;应用ELISA试剂盒检测小鼠血清中肿瘤坏死因子-α(TNF-α)的含量。结果:CⅡ-3对小鼠S180肉瘤的生长具有明显的抑制作用,高、中、低剂量组的抑瘤率分别为58.22%,45.66%,35.37%;CⅡ-3对小鼠的免疫器官具有保护作用,高、中、低剂量组荷瘤小鼠的肝脏指数分别为53.54,54.15,59.53 mg·g-1;脾脏指数分别为3.60,4.44,5.18 mg·g-1;胸腺指数分别为2.45,2.00,2.30 mg·g-1;并能明显提高荷瘤小鼠血清中TNF-α的含量,CⅡ-3高、中、低剂量组TNF-α含量分别为324.92,304.81,278.84 ng·L-1。结论:CⅡ-3能明显抑制小鼠S180肉瘤的生长,对免疫器官没有毒性作用。其提高荷瘤小鼠血清中TNF-α的水平可能是其发挥抗肿瘤作用的途径之一。Objective: Observe the effect of extractive from Periplaneta americana(CⅡ-3) on immune and on inhibitory tumor action in S180-bearing mice.Method: S180 sarcoma was implanted into right-fore oxter,then randomly divided into normal group,model group,cyclophosphamide group(30 mg·kg-1,ip quaque die alterna),CⅡ-3 high dosage group,medium dosage group and low dosage group(200,100,50 mg·kg-1,ig),the S180-bearing mice in every group were received corresponding treatment for 10 days.Tumor growth of the mice was observed,immune organs were weighted to calculate the organ index and ELISA was used to test the concentration of vascular tumor necrosis factor-α(TNF-α) in serum.Results: CⅡ-3 inhibited the growth of S180 sarcoma significantly,200,100,50 mg·kg-1 dosages inhibited the tumor weight by 58.22%,45.66%,35.37%.CⅡ-3 had protective function on immune,the liver indexs in high dosage group,medium dosage group and low dosage group were 53.54,54.15 59.53 mg·g-1 respectively,spleen indexs were 3.60,4.44,5.18 mg·g-1 respectively and thymic indexs were 2.45,2.00,2.30 mg·g-1 respectively.And it dramatically increased the concentration of TNF-α in serum,the concentrations of TNF-α of three groups were 324.92,304.81,278.84 ng·L-1.Conclusion:These results suggest that the CⅡ-3 can obviously inhibit the growth of tumor in S180-bearing mice and toxic effects on immune organs have not fpund,one of anti-tumor effects by CⅡ-3 may be mediated by increasing concentrations of TNF-α in serum.
