摘 要:目的建立山西远志药材高效液相色谱指纹图谱的分析方法,为有效控制远志药材的质量奠定基础。方法采用RP-HPLC法,色谱柱为Hypersil C18柱(4.6 mm×250 mm,5μm),乙腈-0.05%磷酸水梯度洗脱,检测波长为318 nm,流速为1.0 ml/min,分析时间为80 min,分析10批山西远志药材的HPLC指纹图谱,并采用"中药色谱指纹图谱相似度评价软件"、SPSS软件和SIMCA软件进行相似度分析、聚类分析和主成分分析。结果方法学考察实验表明所建立的指纹图谱方法稳定性、精密度和重现性均良好。山西不同来源的远志药材相似度大于0.8,聚类分析和主成分分析结果一致。结论山西远志药材的化学组成一致性较好,质量稳定。所建立的远志药材指纹图谱方法灵敏、稳定,可操作性强。Objective To establish the fingerprint of Polygala tenuifolia Willd.cultivated in Shanxi province by reverse phase high-performance liquid chromatography(RP-HPLC) for the further study on the effective quality control of Yuanzhi.Methods The fingerprint of polygalae(Yuanzhi) was built using Hypersil C18 column(200 mm×4.6 mm,5 μm) and acetonitrile/0.05% H3PO4 in gradient as the mobile phase.The flow rate was set at 1.0 ml/min and the detection wavelength was set at 318 nm.The chromatographic fingerprints were analyzed in 10 batches of Yuanzhi collected from Shanxi province using the Similarity Evaluation System of Chromatographic Fingerprint of Traditional Chinese Medicine,SPSS and SIMCA softwares.Results The similarity of fingerprints of samples collected in Shanxi province was more than 0.80.The results of cluster analysis and principal component analysis were consistent.Conclusion The chemical components of Yuanzhi samples from Shanxi province are of good consistency and have the stable qualities.The method of fingerprint for polygalae is accurate,stable and reproductive and can be used as a scientific foundation for the quality control and evaluation of Yuanzhi.
