寿胎丸对大鼠卵巢移植术后凋亡基因Bcl-2、Bax的影响

摘　　要：目的:探讨寿胎丸对大鼠卵巢移植术后凋亡基因Bcl-2、Bax的影响。方法:210只Wistar雌性大鼠随机分为7组,即自体移植模型组、自体移植+寿胎丸组、异体移植模型组、异体移植+寿胎丸组、异体移植+CsA组、异体移植+CsA+寿胎丸组和正常对照组。各组大鼠于移植术后第7天,第14天,第21天,第28天,第35天分批处死,每次6只。取左侧卵巢,石蜡包埋切片,免疫组化测凋亡相关基因Bcl-2、Bax的表达。结果:移植术后与异体移植模型组相比较,异体移植+寿胎丸组、异体移植+CsA组、异体移植+CsA+寿胎丸组大鼠的Bcl-2水平均升高(P<0.05),Bax水平均降低(P<0.05)。结论:寿胎丸能升调Bcl-2表达,降调Bax表达,升调Bcl-2/Bax的比值,从而发挥抑制卵巢细胞凋亡、抗卵巢移植急性排斥反应的作用。Objective：To investigate the impact of Shoutai Wan on the apoptotie genes, Bcl-2 and Bax after the ovarian transplantation of rats. Methods ：210 female Wistar rats were randomly divided into seven groups, autologous transplantation model group, the autologous transplantation plus Shoutai Wan group, allogeneic transplantation model group, allogeneie transplantation plus Shoutai Wan group, allogeneic transplantation plus CsA group, allogeneic transplantation plus CsA plus Shoutai Wan group and normal control group. Rats in each group were sacrificed in batch on the 7^th ,14^th ,21^st ,28^th and 35^th day after the transplantation,6 rats for each time. The left ovary and paraffin-embedded sections were taken and the expressions of apoptosis related genes Bcl-2 and Bax were measured with immuno- histochemistry. Results ： After transplantation, compared with the allogeneic transplantation model group, the levels of Bel-2 of allogeneic transplantation plus Shoutai Wan group, allogeneic transplantation plus CsA group, allogeneie transplantation plus CsA plus Shoutai Wan group increased（ P 〈 0.05 ） ,while the levels of Bax in these groups decreased（ P 〈 0.05 ）. Conclusion ： Shoutai Wan can enhance the expression of Bel-2,1ower the expression of Bax and raise the Bcl-2/Bax ratio,which plays the role of inhibiting ovarian cell apoptosis and resisting acute rejection of ovarian transplantation.
