摘 要:采用有机溶剂热回流法提取黄连中的小檗碱,并使用响应面法对提取过程中的提取液乙醇浓度、提取温度、固液比对提取效果的影响进行了研究;同时采用原子吸收分光光度法测定经过不同处理后黄连饮片中镉的含量和小檗碱含量来研究黄连中重金属镉的去除。结果表明,黄连中小檗碱的最佳提取工艺为53%的乙醇浓度,提取温度62℃,固液比1:11,提取次数2次,每次1 h;重金属镉去除的工艺以3%氨水6 h+3%NH4NO36 h混合处理效果最佳,镉含量为0.037 6 mg·kg-1,小于0.3 mg·kg-1。小檗碱含量为8.0%。The organic solvents refluxing was used to extract berberine from Coptis chinensis Franch., and the extraction condition, including concentration of ethanol, extraction temperature, solid-liquid ratio were chosen as indices in the extraction process by the response surface method. In the same time, atomic absorption spectrophotometry was employed to test the contents of cadmium and berberine in Coptis chinensis Franch. after removal of cadmium with different treatments. The results showed that the best conditions for extraction berberine were using 53% ethanol, extraction at 62℃, with solid/solvent ratio of 1:11, extraction for 1 h and two times. The best treatment was using 3% ammonia for extracting six hours plus 3% NH4NO3 for six hours, and the content of cadmium is 0.037 6 mg-kg-1, less than 0.3 mg/kg, which meets the pharmacopoeia star.Ldards(2010 edition). The content ofberberine is 8.0%.
