摘 要:目的观察富含半胱氨酸的酸性分泌蛋白(secreted protein acidic rich in cysteine,SPARC)在具有2型糖尿病表型的db/db鼠(10周龄)肝脏组织中的表达情况。方法选择10周龄的db/db鼠(C57BL/KSJ)与其同窝野生对照型各6只,采用反转录-聚合酶链反应(RT-PCR)方法检测肝脏组织中SPARC mRNA的表达水平,免疫荧光染色方法检测肝脏组织中SPARC蛋白的表达水平。结果 SPARC在db/db鼠肝脏组织中呈现高表达(36.91±1.91vs 15.31±0.31)(P<0.01)。结论 SPARC在db/db鼠肝脏组织中呈现高表达,其可能与糖尿病有关的脂肪肝病变有关。Objective Recent studies suggested that secreted protein rich in cysteine(SPARC) was related to diabetes and involved in the development of fatty liver.In this study,we investigated SPARC expression in liver tissue of the db/db mice.Methods We selected 6 db/db mice and 6 littermates of ten weeks old.Anesthetizing by urethane(0.4 g/kg),we took liver tissue.RT-PCR was used to detect SPARC mRNA expression in the liver tissue of db/db mice and the littermate controls.And the differential protein expression of SPARC between two groups was analyzed by immunofluorescence.Results The expression levels of SPARC mRNA and protein in liver tissue of db/db mice were higher than those of controls,36.91±1.91 vs 15.31±0.31(P〈0.01).Conclusion SPARC showed higher expression in liver tissue and SPARC may be involved in diabetes and fatty liver.
