摘 要:[目的]探讨黄芪甲苷拮抗过氧化氢所致小鼠成骨细胞MC3T3-E1凋亡及相关机制.[方法]将成骨细胞MC3T3-E1细胞随机分为A组(正常对照组)、B组(过氧化氢组)、C组(过氧化氢+黄芪甲苷组)及D组(过氧化氢+黄芪甲苷+LY294002组).采用流式细胞术Annexin-V和PI双染检测细胞凋亡情况;同时采用Western-blot检测细胞凋亡相关蛋白Bax,AIF,cyto-C表达情况;免疫荧光和Western-blot检测信号通路相关蛋白P-AKT表达情况.[结果]B组及D组的成骨细胞凋亡率和凋亡相关蛋白Bax,AIF,cyto-C均较A组明显增高(P<0.05);C组的细胞凋亡率和凋亡相关蛋白Bax,AIF,cyto-C活性与A组相比较变化不明显(P>0.05).免疫荧光和Western-blot结果均显示C组的P-AKT表达水平明显增加,与A组比较差异具有统计学意义(P<0.01).[结论]黄芪甲苷可拮抗过氧化氢引起的成骨细胞MC3T3-E1的凋亡,PI3k/AKT信号传导通路可能在其中发挥重要的作用.OBJECTIVE To study the effects and related mechanism of astragaloside on antagonis H2O2 induced osteoblastic MC3T3-E1 cells apoptosis.METHODS MC3T3-E1 cells were divided into group A(normal group),B(H2O2 group),C(H2O2+astragaloside group) and D(H2O2+astragaloside+LY294002 group).The apoptosis of the MC3T3-E1 cells was detected by Annexin-V and PI double staining,and the expression of apoptosis related proteins such as Bax,AIF,cyto-C was measured by Western blot,and the expression of signaling pathway related proteins P-AKT was detected by immunofluorescence and Western blot.RESULTS The rate of osteoblast apoptosis and apoptosis related proteins Bax,AIF,cyto-C in group B and D were significantly increased than that in group A(P0.05).The expression of P-AKT in group C was significantly increased than that in group A(P〈0.01).CONCLUSION The astragaloside could resist the apoptosis of osteoblastic MC3T3-E1 cells induced by H2O2,and PI3k/AKT signal transduction pathway may play an important role in it.
