防己黄芪汤对离体家兔心房肌收缩力及心房钠尿肽分泌调节的影响

摘　　要：[目的]观察防己黄芪汤对离体家兔心房肌收缩力及心房钠尿肽分泌的影响.[方法]采用家兔离体搏动心房灌流模型,观察小、中、大剂量(0.1,0.3,1.0g/L)防己黄芪汤对家兔心房肌收缩力及心房钠尿肽分泌量的影响.采用放射性免疫法测定心房钠尿肽含量.[结果]防己黄芪汤显著增加家兔心房每搏输出量和心房搏动压,与给药前相比较差异具有统计学意义(P<0.05),并呈剂量依赖性,同时可显著抑制心房钠尿肽的分泌(P<0.05).β-受体阻断剂普萘洛尔或L-型钙离子通道阻断剂硝苯地平可部分或完全阻断防己黄芪汤对心房收缩力的增强作用,但均不能改变对心房钠尿肽分泌的抑制效应.[结论]防己黄芪汤主要通过影响β-受体及L-型钙离子通道信号传导途径来增强心房收缩力,并对心房钠尿肽的分泌具有抑制作用.OBJECTIVE To study the effects of Fangji Huangqi decoction（FHD） on atrial dynamics and regulation of atrial natriuretic peptide（ANP） secretion in beating rabbit atria.METHODS By using the beating atrial perfusion model of rabbits,the effects of small,middle and large dosages（0.1,0.3,1.0g/L） of FHD on atrial dynamics and regulation of ANP secretion,and the contents of ANP were measured by radioimmunoassay.RESULTS The FHD increased significantly atrial dynamics and cAMP efflux with dose-dependent manner,and the difference was statistical significance as compared with administration of FHD ago（P〈0.05）,and meanwhile the FHD inhibited significantly ANP secretion（P〈0.05）.Furthermore,enhancement effects of atrial contraction of the FHD was partly or completely blocked by propranolol,a non selective β adrenoceptors inhibitor,and nifedipine,an L type Ca2＋ channel inhibitor,but the inhibitory effects of FHD for ANP secretion were not altered by the inhibitors.CONCLUSION The FHD has positive inotropic effects related with activation of β-adrenoceptors-cAMP L-type Ca2＋ channels signaling pathway and restrains ANP secretion in beating rabbit atria.
