摘  要:[目的]采用高效液相色谱法测定传统朝药材万年蒿中东莨菪内酯、槲皮素及木犀草素的含量.[方法]采用Diamonsil C18色谱柱(250.0mm×4.6mm,5μm),流动相为甲醇-250mL/L磷酸(4∶1)水溶液,流动速度为1.0mL/min,柱温为35℃,检测波长为360nm,线性梯度洗脱.[结果]以峰面积(Y)对进样量(X)进行线性回归,东莨菪内酯、槲皮素和木犀草素的回归方程分别为Y=81 267 X+2 396.7(r=0.999 9),Y=41 356 X-5 037.3(r=0.999 7),Y=13 705 X-741.43(r=0.999 1);线性范围分别为0.106~0.742,0.104~0.728,0.024~0.168μg;加样回收率(n=9)分别为101.38%,RSD为1.94%,101.57%,RSD为1.64%,100.89%,RSD为1.72%.[结论]高效液相色谱法的精密度、稳定性、重复性和加样回收率均较好.所建立的高效液相色谱法具有准确、快速等优点,可用于朝药材万年蒿中3种有效组分的含量测定.OBJECTIVE To determine the contents of three effective ingredients in the Artemisia sacrorum Ledeb by reverse phase-high performance liquid chromatography(RP-HPLC).METHODS RP-HPLC analysis was performed on that chromatographic column was a Diamonsil-C18(250.0mm×4.6mm,5μm),the mobile phase was a mixture of methanol-0.4% phosphoric acid aqueous solution in gradient elution,the flow rate was 1.0mL/min,the column tempratrue was 35℃,and the detection wavelength was 360nm.RESULTS The carlibration curves of scopoletin,quercetol,and luteolol were linear in the range of 0.106-0.742,0.104-0.728 and 0.024-0.168μg,and their regressive equations were Y=81267X+2396.7(r=0.9999),Y=41356X-5037.3(r=0.9997),Y=13705X-741.43(r=0.9991) based on their integral area to their concentration,respectively;and their average recovery rate(n=9)was 101.38%(RSD=1.94%),101.57%(RSD=1.64%)and 100.89%(RSD=1.72%).CONCLUSION The method is accurate,sensitivite and reliable,and the three effective ingredients from Artemisia sacrorum Ledeb.can be rapidly and accurately quantified by RP-HPLC.
