摘 要:目的观察内毒素(Lipopolysaccharide,LPS)对内皮细胞(Endochelial Cell。EC)的活性以及胞内氧化低密度脂蛋白(Oxidized Low Density Lipoprotein,Ox—LDL)与氧化高密度脂蛋白(Oxidized High Density Lipoprotein,Ox—HDL)含量变化的影响。方法选择体外培养的内皮细胞(EC)为研究对象,加入不同浓度的内毒素(分别为10ng/mL、100ng/mL、500ng/mL、1000ng/mL)培养24h后收集细胞,用MTT法检测LPS对内皮细胞增殖的影响,并用酶联免疫吸附法(ELISA)检测胞内Ox—LDL与Ox—HDL含量变化。结果不同浓度的内毒素干预内皮细胞,内皮细胞的活性均下降,以1000ng/mL干预组下降了3980%最显著(P〈0.01)。细胞中的Ox—LDL和Ox—HDL含量均上升,以100ng/mL干预组最显著(P〈0.01),均升高了2倍。结论内毒素能导致内皮细胞损伤.使Ox—LDL和Ox—HDL的合成和释放增加,对动脉粥样硬化的发生发展起到一定的促进作用.Objective To observe the activity of endothelial cell and the effect of Lipopolysaccharide on expression of Oxidized Low Density Lipoprotein and Oxidized High Density Lipoproteina. Methods Cultured endothelial cell (EC) were handled with different concentrations of Lipopolysaecharide group(0,10,100,500 and 1000ng/mL) for 24 h..The activity of EC were detected by MTT assay and the change of Ox-LDL and Ox- HDL in each group were detected by. ELISA. Results EC were treated with different concentrations of Lipopolysaccharide.We found that the activity of EC were decreased ,fell 39.8% to 1000ng/mL the intervention group of the most prominent (P〈0.01).Ox-LDL and Ox-HDL content in a cell are increased to 100ng/mL intervention group of the most prominent (P〈0.01), has warmed twice times.Conclusion Lipopolysaccharide cant result in the damage of EC,and increase the synthesis and release of Ox-LDL and Ox-HDL.Therefore it has played a important roll in atherosclerosis of the occurrence and development.
