摘  要:【目的】探讨锌指蛋白A20对刀豆蛋白A(Concanavalin A,ConA)引起的肝损伤的保护作用。【方法】以刀豆蛋白A诱发小鼠肝损伤,制造肝损伤动物模型,采用尾静脉注射A20表达质粒的方法,观察A20转基因处理组和试验对照组肝组织内TNF-α(tumor necrosis factorα)、IFN-γ(interferon-γ)、IL-2(interleukin 2)细胞因子的表达水平以及肝病理损伤的程度。【结果】A20组ConA处理后肝组织TNF-α、IFN-γ、IL-2等细胞因子的mRNA表达水平及蛋白表达水平均显著低于A20对照组。病理学检查结果提示,A20处理组16 h后小鼠肝病理损伤较对照组有减轻(P<0.05)。【结论】A20能够有效抑制ConA引起的小鼠肝组织炎症反应,并减轻肝组织炎症损伤。[Objective] To explore the protective effect of zinc finger protein A20 on the hepatic injury induced by Concanavalin A. [Methods] Concanavalin A was used to induce hepatic injury. The levels of TNF-a ,IFN-γ ,IL-2 were determined by RT-PCR and ELISA after A20 transgenic therapy. The pathological injury of hepatic tissue was analyzed by traditional pathological examination. [ Results ] After the A20 transgene therap, the mRNA and protein levels of TNF-a ,IFN-γ,IL-2 in the hepatic tissues were reduced significantly. It was showed that the hepatic injury of Group A20 was dramatically than the control group. [Conclusion] The inflammatory reaction of hepatic tissue could be inhibited and the lesions of hepatic tissue by ConA can be alleviative by A20.
