摘  要:目的探讨护骨素基因G1181C位点单核苷酸多态性(SNPs)与冠状动脉性心脏病(CAD)及其严重程度的相关性。方法 368例胸痛患者根据冠状动脉造影检查结果分为非CAD组(146名)和CAD组(222例)。CAD组中单支、双支、三支、四支病变分别为72、59、77、14例。CAD组再根据病史分为急性冠状动脉综合征(ACS)组156例和稳定型CAD组66例。采用介质纯化法提取白细胞DNA,聚合酶链反应(PCR)扩增包含G1181C位点的DNA片段,连接酶检测反应(LDR)检测PCR产物,识别多态性位点。结果非CAD组与CAD组间,非CAD组与ACS组间,ACS组与稳定型CAD组间,非CAD组与不同病变冠状动脉数量组间,护骨素基因G1181C的各基因型频率和分布的差异均无统计学意义(P值均>0.05)。结论研究中未发现护骨素基因G1181C位点SNPs多态性与CAD及其严重程度相关。Objective To investigate the correlation of the single nucleotide polymorphisms (SNPs) of osteoprotegerin gene G1181C with coronary artery disease (CAD) and its severity. Methods A total of 368 patients with chest pain were enrolled in CAD group (n=222) and non-CAD group (n=146), respectively, according to the results of coronary arteriongraphy. Of the 222 CAD patients, 72 had one-vessel, 59 had two-vessel, 77 had three-vessel, and 14 had four-vessel diseases. The CAD patients were subdivided into acute coronary syndrome (ACS, n=156) and stable CAD (n=66) groups. The genomic DNA was extracted by dielectric purification assay. The gene fragment containing G1181C was amplified by polymerase chain reaction (PCR), and then was detected by ligase detection reaction (LDR). Results There were no statistical diferences in genotype frequencies and gene distribution of G1181C between CAD group and non-CAD group, non-CAD group and ACS group, ACS group and stable CAD group, and non-CAD people and patients with different vessel diseases. Conclusion The osteoprotegerin gene G1181C polymorphisms are not associated with the presence and severity of CAD in Chinese Han people in Shanghai area.
