摘　　要：目的探讨组织蛋白酶B(Cathepsin B Cat B)在大鼠酒精性肝病中的表达及其意义。方法酒精灌胃法制备大鼠酒精性肝病模型。检测大鼠肝指数及血清天门冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)含量、HE染色观察肝脏病理学改变,逆转录聚合酶链反应(RT-PCR)、免疫组化技术检测肝组织中Cat B mRNA水平及蛋白表达的变化。结果模型组肝细胞损伤明显,肝指数模型组较正常组显著升高(P﹤0.01),模型组血清ALT和AST较正常组明显升高(P﹤0.05);RT-PCR及免疫组化均显示,模型组肝组织中CatB表达较正常组显著上调(P﹤0.01)。结论 Cat B可能参与了酒精性肝病的发生、发展过程,并推测其在酒精性肝病肝细胞凋亡过程中可能发挥了重要的作用。OBJECTIVE To investigate the expression and significance of Cathepsin B（Cat B）in alcoholic liver disease in rat.METHODS Alcoholic liver disease in rats were induced by gavages.Liver coefficient of the rats were calculated.The levels of serum aspartate aminotransferase（AST）,alanine aminotransferase（ALT）were measured.The pathological changes of liver were observed after H-E.RT-PCR and immunohistochemistry were used to detect the change of Cat B protein expression and mRNA in alcoholic liver disease in rat.RESULTS Compared with that in the control group,liver coefficient of the rats increased（P﹤0.01）.LM observed that the hepatocytes were obviously injured.The levels of serum AST and ALT significantly increased in the model group（P﹤0.05）compared with that in the control group.CatB expression in the liver determined by immunohistochemistry and RT-PCR significantly increased in the model group（P﹤0.01）.CONCLUSION It is presumed that Cat B participates in the genesis and development of ALD,and it may play an important role in the hepatocytes apoptosis.
