银屑病患者单核细胞诱生的郎汉斯细胞特征分析

摘　　要：目的　从郎汉斯细胞 (LC)的分化发育过程探讨其在银屑病患者体内异常的原因。方法　将外周血单核细胞经GM CSF +IL 4+TGF β1联合培养 5d ,分别通过流式细胞仪、混合淋巴细胞反应分析其表型及其抗原提呈功能 ,并通过ELISA法检测其分泌功能。结果　银屑病患者单核细胞经GM CSF +IL 4+TGF β1联合培养后能分化发育为具有LC相对特征性抗原标志CDla的细胞 ,其比例、表型特征、胞内合成及分泌IL 1 2的能力、刺激T淋巴细胞的活性等与健康对照组相比差异无显著性。结论　银屑病患者LC的异常并非发生于其前体细胞 。Objective To investigate the mechanism of abnormality of LCs in patients with psoriasis after analysis of the characteristics of LC derived from the monocytes. Methods The monocytes isolated from the peripheral blood of patients with psoriasis and healthy blood donors were cultured with GM CSF+IL 4+TGF β1 for 5 days. Their phenotypes, antigen presenting ability and the IL 12 secreted by the cells were analyzed by FACS caliber,MLR and ELISA.Results After cultured with GM CSF+IL TGF β1,LCs which expressed CDla were induced from the monocytes not only from healthy blood donors but also from patients with psoriasis. The proportion of CDla + cells, the phenotypes, the stimulatory capacity and the IL 12 secreted by the cells were the same in patients with psoriasis and healthy blood donors. Conclusions The monocytes in patients with psoriasis have the capacity to differentiate into LCs and the abnormalities of which were not due to their progenitors. The data suggested that the abnormalities of LCs in patients with psoriasis might already exist in their microenvironment before entering the skin tissue.(Shanghai Med J, 2000,23∶138 141)
