龙力胶囊对人肝癌细胞诱导分化及机理研究

摘　　要：目的 :研究中药复方龙力胶囊对人肝癌HCC 92 0 4的诱导分化作用及其机理。方法 :用体外培养方法培养HCC 92 0 4细胞 ,观察龙力胶囊对HCC 92 0 4细胞形态学、集落形成能力、AFP与ALB分泌量、γ GT活力、核型、细胞周期以及癌基因表达的影响。结果 :龙力胶囊能改善HCC 92 0 4细胞的异型性 ,降低其集落形成能力 ,降低AFP分泌量及γ GT活力 ,提高ALB分泌量 ,降低细胞染色体主流范围 ,降低c myc、c H ras基因表达 ,但对细胞周期影响不大。结论 :龙力胶囊具有诱导HCC 92 0 4细胞分化的作用 ,其机理可能涉及核型良性变及c myc、c H ras降调节。Objective: To investigate the effects and meachanism of Chinese medicine compound/dragon power capsule (DPC) induced differentiation on human hepatocarcinoma cell lines Hcc 9240. Methods: The present study was carried out by using technique of cell culture in vitro. The effect was observed by monitoring cytomorphology, cloney forming, the secretion of AFP and ALB,γ GT activity,chromosome,cell cycle and expression of oncogene. Results:Morphology of HCC 9204 became well differentiated, growth became slower, cloney forming was inhibited, the secretion of AFP and γ GT activity were reduced, the secretion of ALB increased and expression of c myc. C H ras oncogene decreased, the main range of chromosome reduced, but cell cycle did not change markedly. Conclusions: DPC had the effect of induced differentiation on HCC 9204, the mechanism might be related to chromosome and Down Regulation of c myc. c H ras oncogene.
