摘  要:目的探究组织工程水凝胶纤维丝的成形工艺方法,分析成形参数对纤维丝成形的影响。方法利用海藻酸钠在二价阳离子作用下迅速交联的机制,采用海藻酸钠基材料在氯化钙溶液中连续挤出成丝的方法制备纤维丝,并在旋转流场中完成纤维丝的缠绕收集。结果在合适的成形参数下,实现水凝胶纤维丝的连续稳定成形,得到直径小且均匀的水凝胶纤维丝,还制备了含有细胞的水凝胶纤维丝,结果表明细胞在丝中能够有效存活,生长情况良好。结论基于微单元体组装的"自下而上"组织工程构建思路,成功设计开发了纤维丝的成形工艺,系统研究了成形质量与成形参数之间的关系,成功制备了纤细均匀的和含有细胞的水凝胶纤维丝。Objective To develop the forming process of tissue engineered hydrogel fibers and investigate the impact of parameters on the forming. Methods Utilizing the mechanism that sodium alginate can be rapidly cross-linked under the action of bivalent cations, hydrogel fibers were produced and collected in winding shape continuously extruding alginate-based materials inside the rotating CaC12 solution. Results Hydrogel fibers were formed continuously and steadily with the optimized parameters and the slender fibers with regular shape and uniform diameter were obtained. In addition, the hydrogel fibers containing cells were also successfully prepared. The results that cells can survive effectively and grow well in the fibers. Conclusion Based on the "bottom-up" approach in tissue-engineering construction through modular assembly, the forming process of hy- drogel fibers has been designed and developed successfully in this paper. The relationship between the forming result and the parameters is studied systematically and two kinds of hydrogel fibers, that is of good shape and containing cells, are created.
