高效液相色谱法测定穿虎痛风合剂中大黄素含量

摘　　要：目的建立测定穿虎痛风合剂中大黄素含量的高效液相色谱法。方法采用Agilent Zorbax C18柱(250 mm×4.6 mm,5μm),以甲醇-0.1%磷酸溶液(80∶20)为流动相,柱温为35℃,流速为1.0 mL/min,检测波长为254 nm。结果大黄素进样量在5.742～86.13μg范围内与峰面积呈良好线性关系(r=0.999 9),平均加样回收率为98.95%,RSD=0.99%(n=9)。结论所建立的方法简便可靠、分离度好,可用于穿虎痛风合剂的质量控制。Objective To establish a method for the determination of emodin in Chuanhu Tongfeng Mixture.Methods The separation was performed on the Agilent Zorbax C18 column（250 mm×4.6 mm,5 μm）.The mobile phase was methanol-0.1%H3PO4（80：20） with the flow rate of 1.0 mL/min;The detection wavelength was 254 nm and the column temperature was 35 ℃.Results Under established the chromatographic condition,the calibration curve showed the good linearity in the range of 0.086 13-0.430 6 μg for emodin sample size（r=0.999 9）.The average recovery rate was 98.95%,RSD=0.99%.Conclusion The method is convenient and reliable with good separation,which could be used for the quality control of Chuanhu Tongfeng Mixture.
